摘要
对弱碱性阴离子交换树脂分离纯化海带岩藻聚糖硫酸酯的工艺条件进行了研究,并采用苯酚-硫酸法测定总糖浓度绘制解吸曲线,确定最佳样液的pH值和洗脱流速。结果表明:在上样液pH为7.0、洗脱流速为1.0 mL/min时,分别收集0.5 mol/L和1.0 mol/L的NaCl溶液的洗脱部分,得到F-1、F-2两个组分,其回收率分别为22.5%、10.6%,总糖的质量分数分别为30.5%、28.6%。其中F-1为低硫组分,硫酸根的质量分数为11.7%,F-2为高硫组分,硫酸根的质量分数为26.5%
Weaky basic anion - exchange resin was used to isolate and purify fucoidan from kelp Laminaria japonica, and phenol -H2SO4 method was applied to determine total carbohydrates and the optimal processing condition. Under the conditions of sample solution with pH value of 7.0, and elution rate of 1.5 mL/min, F - 1, F - 2 fractions were obtained by collecting the solution with 0. 5 mol/L and 1.0 mol/L of NaCI elution respectively, the yields were 22. 5% and 10. 6% ,the total sugar contents were 30. 5% ,28.6%, and the contents of SO4^2- were 11.7% and 26. 5 %. It can be concluded that weakly basic anion - exchange resin is suitable for isolation and purification of the kelp fucoidans.
