摘要
采用正交试验的方法,研究了用戊聚糖复合酶与复合纤维素酶酶解海带Lam inaria japonica提取岩藻聚糖硫酸酯的工艺参数。结果表明:用戊聚糖复合酶酶解海带的最佳酶解参数为酶加量0.06%,温度为40℃,pH为3.5,酶解时间为30 m in,对岩藻聚糖硫酸酯的提取率为1.845%,粗提物中总硫酸根的质量分数为0.238%;用复合纤维素酶酶解海带的最佳酶解参数为酶加量0.208%,温度为50℃,pH为4.5,酶解时间为50 m in,对岩藻聚糖硫酸酯的提取率为1.279%,粗提物中总硫酸根含量为0.231%。用两种复合酶酶解法提取的岩藻聚糖硫酸酯粗提物提取率均较水煮法高(0.968%)。
The enzymatic hydrolytic technologies of fucoidan from kelp Laminaria japonica were studied by Novozymes Viscozyme L and Celluclast BG. An orthogonal test was designed to evaluate the optimum factors for Novozymes Viscozyme L hydrolysis. It was found that the optimal conditions were as the following: 0.06% Novozymes Viscozyme L, at temperature of 40℃ pH 3.5 and 30 minutes when 1. 845% of yield rate of fucoidan extract was obtained, in which the content of SO^2-4 was 0.238%. The optimal factors for Celluclast BG were the following: 0.208% Celluclast BG, at temperature of 50%, pH 4.5 and 50 minutes when 1. 279% of yield rate of fucoidan extract was achieved, in which the content of SO^2-4 was 0.231%. There was higher yield rate of fucoidan by enzymatic hydrolysis than by the traditional hydrolysis(0.968% ).
