摘要
采用两肾一夹型建立肾血管性高血压大鼠模型。探讨岩藻聚糖硫酸酯低聚糖对肾血管性高血压大鼠的血压、呼吸及血浆中血管紧张素 (Ang )含量的影响。用 GY- 6 0 88型多道生理参数分析记录仪和张力传感器测定、记录血压和呼吸 ;用均相竞争法测定 Ang 的含量。实验表明 ,1 2 .5 ,2 5 ,5 0 mg/ kg的岩藻聚糖硫酸酯低聚糖均能显著地降低肾血管性高血压大鼠的动脉血压。其中 ,高剂量 (5 0 mg/kg)的降压效果与 1 4 mg/kg的卡托普利相当 ;给药大鼠的呼吸频率随血压的变化呈正相关变化 ;1 2 .5 ,2 5 ,5 0 mg/ kg剂量的岩藻聚糖硫酸酯低聚糖均能降低高血压大鼠血浆中 Ang 的含量。其中 ,高剂量 (2 5 ,5 0 mg/ kg)组的 Ang 水平同卡托普利组 (1 4 mg/ kg)相当。岩藻聚糖硫酸酯低聚糖对肾血管性高血压大鼠有显著的降压作用 ,抑制 Ang 生成可能是其降压机制之一。
In this paper, the antihypertensive effects of fucoidan oligosaccharides prepared from Laminaria japonica on renovascular hypertensive rats (RHR) and related mechanisms are presented. Renovascular hypertensive rats (RHR) models were made with a two kidney one clip assay and the effects of the fucoidan oligosaccharides at doses of 6.25,12.5, 25 and 50mg/kg were studied in renovascular hypertensive rats in the acute therapeutic experiment. Arterial blood pressure including systolic blood pressure (SBP) and diastolic blood pressure (DBP), respiration and plasma angiorensinⅡ(AngⅡ) were measured respectively. The results show that fucoidan oligosaccharides can markedly reduce the arterial blood pressure and plasma AngⅡ of RHR and the antihypertensive effects of fucoidan oligosaccharides at high doses are similar to captopril. Moreover, the change of the respiration of RHR is similar to that of their blood pressure. The respiration of RHR had the same change trend as blood pressure. These results indicate that fucoidan oligosaccharides have good antihypertensive effects on RHR and one of the mechanisms underlying the antihypertensive effects might be that they can inhibit the production of AngⅡ.
