摘要
以2个分离自云南鸡足山的野生离褶伞菌株C821-001和C822-003为试材,根据ITS序列分析,对2个野生菌株进行分类鉴定。采用液体摇瓶培养法,以菌丝生物量为主要指标,对C821-001和C822-003进行液体培养基筛选,从5种不同培养基中分别筛选出适合2个菌株菌丝生长的液体培养基。结果表明:经ITS序列分析证实C821-001为真姬离褶伞Lyophyllum shimeji,C822-003为烟熏褐离褶伞Lyophyllum infumatum。C821-001和C822-003均在配方5中生长最好,菌丝体干质量分别达到1.948 8 g和1.128 8 g,因此,配方5可作为2个菌株菌丝体繁殖的首选液体培养基。
Two wild strains,named C821-001 and C822-003,were isolated from two spieces of Lyophyllum spp.which were collected in Jizushan of Yunnan Province,then these two selected samples were identified by ITS sequence analysis.The liquid culture medium of C821-001 and C822-003 were selected by liquid shaking flask culture method with mycelial biomass as the main index,the liquid culture medium suitable for mycelial growth of two selected strains were selected from 5 kinds of culture medium which consisted of different materials.The results showed that C821-001 and C822-003 were respectively identified as Lyophyllum shimeji and Lyophyllum infumatum analyzed by phylogenetic relationship of ITS sequences.Both C821-001 and C822-003 grew best in formula 5,with the dry weight of mycelium 1.948 8 g and 1.128 8 g,respectively.So the formula 5 could be used as the preferred liquid culture medium for mycelium propagation of two selected strains.
