摘要 抗体是实验研究和药物应用的重要工具。大多数抗体具有4条多肽链的对称结构,两条较长的重链和两条较短的轻链,两臂末端是相同的抗原结合部位。除了这些传统的抗体,羊驼、一些骆驼和鲨鱼体内会产生一种只含有重链的抗体,称为重链抗体。它们的抗原结合部位只由单一结构域构成。这类单域抗体具有分子小、可溶性好、热稳定性高和体内组织渗透性好等优点,引起了药物开发与免疫治疗应用等领域的广泛关注,具有很广阔的研究前景。文章主要概述了抗体药物的研究,单域抗体的特点及天然物种来源的单域抗体的研究概况。 Antibodies are important tools for experimental research and drug applications. Most antibodies have a symmetric structure of four polypeptide chains,two longer heavy chains( H chains) and two shorter light chains( L chains),and both arm ends are the same antigen binding sites. In addition to these traditional antibodies,the bodies of alpacas,some camels,and sharks could produce an antibody that contains only heavy chains,called heavy chain antibodies. Their antigen binding sites only consist of a single domain. These single domain antibodies have the advantages of small molecules,good solubility,high thermal stability and good tissue permeability in vivo. It has aroused widespread concern in the fields of drug development and immunotherapy applications,and has a wide range of research prospects. The research of antibody drugs,the characteristics of single-domain antibodies and the research profile of single-domain antibodies derived from natural species are mainly summarized.
