慢性束缚应激对小鼠攻击行为的影响及机制探讨

摘要 [目的]研究慢性束缚应激对小鼠攻击行为的影响及其机制。[方法]选取52只7周龄C57BL/6J雄性小鼠,随机分为对照组(control group,Con)和慢性束缚应激组(chronic restraint stress group,RS)。另选取7周龄24只小鼠(公母各半),配对饲养,同样分为2组(对照组和慢性束缚应激组),其中雄性小鼠作为驻留者用于攻击行为评测,雌性小鼠仅用作陪伴。再选取6周龄雄性小鼠12只作为入侵者用于攻击行为评测。慢性束缚应激组小鼠每天束缚6 h(09:00—15:00),连续处理7 d,应激小鼠处理期间对照组小鼠禁食禁水,而雌性小鼠和入侵者小鼠未做任何处理。记录试验起始及结束时小鼠体重,检测小鼠攻击行为、血浆和下丘脑神经递质水平、下丘脑攻击行为相关基因及蛋白表达量。[结果]与对照组相比,慢性束缚应激极显著抑制小鼠生长(P<0.01),显著降低小鼠攻击潜伏期时间(P<0.05),显著增加小鼠总攻击次数(P<0.05),显著上调小鼠下丘脑囊泡单胺类转运体2、5-羟色胺受体1b、乙醛脱氢酶1a1、多巴胺脱羧酶、多巴胺受体2及多巴胺转运体mRNA水平(P<0.05或P<0.01),极显著下调小鼠下丘脑DAT蛋白表达量(P<0.01)。[结论]慢性束缚应激可降低小鼠生长性能,增加小鼠攻击行为,攻击行为的改变可能与下丘脑DAT蛋白低表达密切相关。 [Objectives]The paper aimed to study the effects of chronic restraint stress on the aggressive behavior and mechanism of mice.[Methods]Fifty-two 7-week-old C57BL/6J male mice were selected and randomly divided into control group(Con)and chronic restraint stress group(RS),respectively.Another twenty four 7-week old mice(male and female in half)were randomly selected,reared in pairs and divided into two groups as follows.Among them,male mice were used as resident for aggression evaluation,female mice were only used as companions.In addition,twelve 6-week-old male mice were selected as intruders for aggression evaluation.The mice in the chronic restraint stress group were treated for 7 days and restrained for 6 h every day(09:00-15:00).The mice in the control group were fasted and dehydrated during this period,and the female mice were not treated.Seven days after treatment,twelve male mice at 6 weeks of age were selected as intruders for the aggressive behavior test.The initial weight and final weight of mice were recorded.The aggressive behavior,plasma and hypothalamic neurotransmitters levels,and the expression levels of genes and proteins related to the aggressive behavior were examined in mice.[Results]Compared with the control group,chronic restraint stress could significantly inhibit the growth performance(P<0.01),reduce attack latency time(P<0.05),increase total number of attacks(P<0.05),enhance the levels of hypothalamic vesicular monoamine transporter 2,5-hydroxytryptamine 1b receptor,acetaldehyde dehydrogenase 1a1,dopa decarboxylase,dopamine receptor 2 and dopamine transporter mRNA(P<0.05 or P<0.01),and decrease the expression level of hypothalamic DAT protein in mice(P<0.01).[Conclusions]Chronic restraint stress could impair growth performance and increase aggressive behavior,which is closely related to the lowered hypothalamic DAT protein expression in mice.
