安脑平冲方对脑出血大鼠脑血肿周围组织Tf及TfR的影响 被引量:9

Effect of Annaopingchong Decoction on Expression of Transferrin and Transferrin Receptor in The Tissues Around Cerebral Hematoma of Rats with Intracerebral Hemorrhage

摘要 目的观察安脑平冲方对脑出血模型大鼠神经功能缺损、脑组织铁沉积及脑血肿周围组织转铁蛋白(Tf)、转铁蛋白受体(TfR)表达水平的影响。方法将240只雄性SD大鼠随机分为假手术组、模型组、安脑平冲方低剂量组、安脑平冲方中剂量组、安脑平冲方高剂量组、吡拉西坦组,每组再分为6 h、3 d、7 d、14 d 4个亚组,每个亚组10只大鼠。使用大鼠自体全血于左侧尾状核立体定位注射,并采用二次注血退针法复制脑出血大鼠模型。造模成功后的大鼠,除假手术组和模型组外,均给予相应药物进行干预,并对各组各时间点的动物采用Berderson法进行神经功能缺损评分,摩列利铁染色法检测脑组织铁沉积的变化,Western blotting法检测脑血肿周围组织Tf和TfR的蛋白表达水平。结果模型组及各药物组不同时间点的神经功能缺损评分均高于假手术组(P<0.01);安脑平冲方低、中、高剂量组7、14 d的神经功能缺损评分均低于模型组(P<0.05)。铁沉积平均光密度值统计分析结果显示模型组6 h可见铁离子沉积,3 d逐渐升高,第7日达峰值,第14日有所降低;模型组和各药物组在不同时间点的铁沉积密度值均显著高于假手术组(P<0.01),各药物组在3、7、14 d的铁沉积平均光密度值与同期模型组比较下降显著(P<0.01)。Western blotting检测结果表明,模型组各时间点Tf和TfR蛋白表达较假手术组明显升高(P<0.01),安脑平冲方低、中、高剂量组各时间点Tf和TfR蛋白表达较模型组降低(P<0.05或P<0.01)。结论安脑平冲方能改善脑出血大鼠的神经功能损伤,其机制可能为通过下调Tf和TfR来减少铁离子向神经细胞中转移,从而产生神经保护作用。 Objective:To observe the effect of Annaopingchong Decoction on the neurological deficit,brain iron deposition and the expression of Tf and TfR in cerebral hematoma rats.Methods:240 male rats were divided into sham-operated group,the model group,low-dose group,medium-dose group,high-dose group and piracetam group.Each group was divided into four subgroups of 6 h,3 d,7 d and 14 d,with 10 rats in each subgroup.ICH model was established by stereotactic injecting autologous whole blood into the left caudate nucleus and withdrawing needle twice.After successful modeling,rats were intervened with corresponding drugs.Berderson neurological deficit score and Mallouy iron staining were used to detect the changes of neurological deficit and iron deposition in brain tissue at corresponding time points.Western blotting was used to detect the expression of Tf and TfR in the tissues around cerebral hematoma.Results:The neurological deficit scores of the model group and each therapy group at different time points were higher than those of the sham-operated group(P<0.01).And the neurological deficit scores of the low,medium and high dose groups were decreased on 7 d and 14 d(P<0.05).Iron deposition density analysis showed that iron deposition was observed in the model group at 6 h,and gradually increased at 3 d,peaked at 7 d,and decreased at 14 d.And the iron deposition of the model group and each therapy group were significantly higher than that of the sham-operated group at different time points(P<0.01).The iron deposition of the therapy groups at 3 d,7 d and 14 d were lower than those of the model group(P<0.01).The results of Western blotting showed that the expression of Tf and TfR in the model group was significantly higher at each time point than that in the sham-operated group(P<0.01).And that in the low,medium and high dose groups were lower at each time point than those in the model group(P<0.05).Conclusion:Annaopingchong Decoction protects against ICH injury by improving neurologic deficits,reducing the transfer of iron
