锌指蛋白A20在大鼠脑出血后脑、肺组织中的表达及相关研究 被引量:1

Expression of Zinc Finger Protein A20 in Brain and Lung Tissues and Its Role after Intracerebral Hemorrhage of Rats

摘要 目的探讨脑出血后锌指蛋白A20(A20)在脑和肺组织的表达以及在急性肺损伤中的作用。方法通过自体血尾状核注入法构建脑出血大鼠模型。雄性SD大鼠随机分为假手术对照组(n=12)和脑出血组(n=72),脑出血组再按时间点分为3、6、24、48、72和168 h6个亚组(均n=12)o测定肺水肿、支气管肺泡灌洗液(BALF)总蛋白浓度,苏木精-伊红染色观察脑出血后脑组织和肺组织的周围炎症反应情况,免疫组化‘Western blot检测脑、肺组织中A20、核转录因子-kB(NF-kB)的表达。结果脑岀血后3 h出现肺水肿、BALF总蛋白浓度增加,48 h达到高峰(P<0.05);脑血肿周围组织72 h A20、NF-kB表达量达高峰(P<0.05);肺组织48 h A20、NF-kB表达量达高峰(P<0.05)o结论A20在脑出血后脑组织和肺组织中表达均有升高,提示A20可能在脑出血后急性肺损伤的发生发展过程中发挥作用。 Aim To investigate the expression of zinc finger protein A20 in brain and lung tissues after intracerebral hemorrhage(ICH)and its role in acute lung injury.Methods A rat model of ICH was constructed by autologous blood injected into caudate nucleus.Rats were randomly divided into a sham group(12 rats)and a ICH group(72 rats).The ICH group was divided into 6 subgroups of 3 h,6 h,24 h,48 h,72 h and 168 h(12 rats in each subgroup).The pulmonary edema and the total protein concentration of bronchoalveolar lavage(BALF)were measured.The inflammatory reaction surrounded the brain and lung tissues after ICH was examined by HE staining.Immunohistochemistry,Western blotting were used to detect A20,NF-kB in brain and lung tissues.Results Pulmonary edema and total BALF protein concentration increased at 3 h after ICH,and reached a peak at 48 h(P<0.05)・In comparison,the peak of the expression about A20,NF・kB in brain tissue was at 72 h(P<0.05)while the peak in lung tissue was at 48 h(P<0.05)・Conclusion The expression of A20 in brain and lung tissue were increased after ICH,suggesting A20 could be a potential role in the development of acute lung injury after ICH.
