摘要
目的观察奥拉西坦对大鼠急性脑出血(ICH)血肿周围Caspase-3表达的影响,探讨奥拉西坦对脑出血大脑的保护作用。方法采用立体定位钻孔自体血注入法制备大鼠内囊出血模型,对照组只切开头皮,不做任何处理;模型组及奥拉西坦高、低剂量组分别给予相应药物腹腔注射,在6 h、12 h、24 h 3个时间点分别取材,通过免疫组化的方法检测Caspase-3蛋白的表达变化。结果手术组的大鼠较对照组Caspase-3蛋白的表达明显增多,其中奥拉西坦高、低剂量组Caspase-3蛋白表达较模型组明显减少(P均<0.01),奥拉西坦高、低剂量组Caspase-3蛋白表达比较有显著性差异(P均<0.05)。结论奥拉西坦可通过调节Caspcase-3免疫阳性细胞的表达,抑制脑出血周围神经细胞凋亡,促进细胞代谢,从而对脑出血大鼠大脑起保护作用。
Objective It is to observe the influence of Oxiracetam on expression of Caspase - 3 in nerve cells of rat with a- cute cerebral hemorrhage (ICH) around the hematoma, to explore its neuroprotective mechanism. Methods Using three -di- mensional localization autologous blood injection method to establish rat models with internal capsule hemorrhage, the rats" scalp of control group was only cut open without any treatment. Model group and high and low dosage of Oxiracetam groups were treated with corresponding drugs by intraperitoneal injection. The brain tissue was gotten at 6 h, 12 h, 24 h to detect the expression of Caspase - 3 by immunohistochemistry. Results Compared with the control group, Caspase - 3 protein expression was significantly increased in the surgery group. The expression of Caspase - 3 protein was obviously reduced in high and low dosage of Oxiracetam groups compared with the model group (P 〈 0.01 ). The difference in the expression of Caspase - 3 be- tween high dose Oxiracetam group and low dose Oxiracetam group was significant (P 〈 0.05 ). Conclusion Oxiracetam can regulate the Caspcase- 3 immune cells'expression, inhibit neural cell apoptosis around cerebral hemorrhage and promote cell metabolism, thus to play a protecting role on brain in rats with cerebral hemorrhage.
