摘要
目的:探讨黄芩素对C57BL/6小鼠代谢综合征的可能保护作用。方法:10周龄的雄性C57BL/6小鼠(n=120),随机数字表法分成4组:HFD(high fat diet group)组、HFD+0.5%Bacalein组、NC组、NC+0.5%Bacalein组。在实验期间,每周称量记录小鼠食量;在实验开始6周内,每周均测量记录小鼠体重,以后每两周称量一次小鼠体重。在实验第23,24周,分别行小鼠腹腔注射葡萄糖耐量试验(IPGTT)、腹腔注射胰岛素耐量试验(IPITT),计算曲线下面积(AUC)。在实验第29周末终点取材,检测血脂,处死小鼠,快速分离出小鼠全部肝脏、白色脂肪组织,称重并计算出肝脏指数、脂肪指数。结果:从实验第2,5周开始,HFD组小鼠体重分别高于NC组(P<0.05)和HFD+0.5%Bacalein组小鼠体重(P<0.05)。空腹血糖,HFD组与HFD+0.5%Bacalein组无明显差异(P>0.05),IPGTT、IPITT及各自曲线下面积(AUC),HFD组与HFD+0.5%Bacalein组小鼠差异显著(P<0.001)。TC、LDL-C检测,HFD组与HFD+0.5%Bacalein组之间比较(P<0.01)。肝脏组织学观察显示,HFD组小鼠肝脏细胞脂肪空泡变性显著,空泡融合成大片状,而高脂饲料加黄芩素干预后,肝细胞脂肪空泡变性明显减少,肝小叶清晰。结论:成功建立C57BL/6小鼠代谢综合征疾病模型,黄芩素有效改善胰岛素抵抗,调节糖耐量,降低TC和LDL-C,对高脂引起的肝脏脂肪变性有显著保护作用。
Objective: To investigate the possible protective effect of baicalein on C57 BL/6 mice with metabolicsyndrome. Methods: One hundred and twenty male C57 BL/6 mice at 10 weeks of age were divided into four groupsusing the random number table method: high-fat diet(HFD) group, HFD + 0.5% baicalein group, normal control(NC) group, and NC+0.5% baicalein group. During the experiment, the eating capacity of the mice was measured ev-ery week. The weight of the mice was recorded every week during the first 6 weeks and then every two weeks for thefollowing experiment period. At weeks 23 and 24, the intraperitoneal glucose tolerance test(IPGTT) and intraperito-neal insulin tolerance test(IPITT) were conducted, and the area under the receiver operating characteristic curve(AUC) was calculated. At the end of the 29 th week, blood lipids were determined, and then the mice were sacrificedto quickly obtain the liver and white adipose tissue for measuring the weight and calculating the liver index and fatindex. Results: Starting from the 2 nd and 5 th weeks, the weight of the mice in the HFD group significantly exceededthat of the NC group and HFD + 0.5% baicalein group, respectively(P < 0.05). Fasting blood glucose showed a sig-nificant difference between the HFD group and the HFD + 0.5% baicalein group(P > 0.05). There were significantdifferences in the results of IPGTT and IPITT and their AUCs between the HFD group and the HFD+0.5% baicaleingroup(P < 0.001). There were significant differences in total cholesterol(TC) and low-density lipoprotein cholesterol(LDL-C) between the HFD group and the HFD + 0.5% baicalein group(P < 0.01). Liver histology showed significant fatty vacuolation of hepatocytes in the HFD group, and the vacuoles fused into large ones;baicalein significantly re-duced fatty vacuoles and made the hepatic lobules more clear in the mice fed with HFD. Conclusion: We success-fully establish a C57 BL/6 mouse model of metabolic syndrome, and find that baicalein effectively improves insulinresistance, regulates glucose
