柴胡、黄芩组方合剂对糖尿病肾病大鼠尿蛋白、脂质代谢的影响 被引量:10

摘要 目的:观察柴胡、黄芩组方合剂对糖尿病肾病大鼠尿蛋白、脂质代谢的影响。方法:实验于2003-06/12在解放军第一军医大学中医系完成。将32只Wistar大鼠随机分为模型对照组、大剂量药物治疗组、中剂量药物治疗组和小剂量药物治疗组,每组8只。用链脲佐菌素腹腔注射法诱导大鼠制备糖尿病肾病模型,模型对照组用生理盐水灌胃,小剂量药物治疗组用1.8g/kg柴胡、黄芩组方合剂灌胃,大、中剂量组用药剂量分别是小剂量药物的4倍和2倍。治疗12周后检测大鼠血糖、24h尿蛋白、24h尿白蛋白排泄率、胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇的含量变化。柴胡、黄芩组方合剂为广州一方制药厂生产的,小四五颗粒主要成份:柴胡、黄芩、半夏、人参、白术等。结果:32只大鼠纳入结果分析。①小、中、大剂量药物治疗组24h尿蛋白定量分别为(56.20±11.30,44.22±10.68,39.22±18.44)mg/24h,模型对照组为(64.74±16.34)mg/24h,中剂量和大剂量药物治疗组的24h尿蛋白定量比模型对照组低(P均<0.05),小剂量药物治疗组与大剂量药物治疗组之间差异有显著性意义(P<0.05)。②各剂量组24h尿白蛋白定量分别为(0.27±0.43,1.07±0.26,1.08±0.29)mg/24h,均显著低于糖尿病模型对照组(1.84±0.74)mg/24h(P均<0.05),各剂量组之间差异不明显。③各剂量组和模型对照组胆固醇含量分别为(1.48±0.15,1.21±0.12,1.13±0.12,1.19±0.11)mmol/L,三酰甘油分别为(1.10±0.27,0.74±0.17,0.65±0.13,0.70±0.12)mmol/L,各剂量组均显著低于对照组(P<0.05),但各剂量组间比较差异不显著。结论:柴胡、黄芩组方合剂可以降低糖尿病肾病大鼠尿蛋白、白蛋白排泄,对其脂质代谢也具有调节作用。 AIM:To observe effect of mixture of chaihu and huangqin on urine protein and lipid metabolism in rats with diabetic nephropathy. METHODS:The experiment was completed in the Department of Traditional Chinese Medicine, the First Military Medical University of Chinese PLA from June to December 2003.Totally 32 Wistar rats were divided randomly into model control group, high-dosage group, middledosage group and low-dosage group with 8 in each group.Rats were injected intravenously with streptozotocin (STZ) to establish models of diabetic nephropathy,Rats were perfused with saline in model control group, with 1.8 g/kg mixture of chaihu and huartgqin in low-dosage group, with 7.2 g/kg and 3.6 g/kg mixture of chaihu and huangqin in high-dosage and middle-dosage groups, respectively. Twelve weeks after treatment, changes of blood glucose, 24-hour urine protein, 24-hour excretory rate of urine albumin, cholesterol, triacylglycerol (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C). Mixture of chaihu and huangqin was provided by Guangzhou Yifang Pharmaceutical Factory, and xloJ〉si-wu granule was consisted of Chinese thorowax root,baikal skullcap root,Pinellia tuher,ginseng,largehead atractylodes rhizome, etc. RESULTS: Totally 32 rats entered the final analysis. ① Contents of 24-hour urine protein were (56.20±11.30, 44.22±10.68, 39.22±18.44) rag/24 hours in low-, middle-and high-dosage groups respectively, and (64.74±16.34) mg/24 hours in model control group. Quantity of 24-hour urine protein was lower in middle-and high-dosage groups than that in model control group (P 〈 0.05), and there was significant difference between low-dosage group and highdosage group (P 〈 0.05), ② Quantities of 24-hour urine albumin were (0.27±0.43, 1.07±0,26, 1.08±0.29) mg/24 hours in low-, middle- and highdosage groups respectively, which were all lower than that (1.84±0.74) mg/ 24 hours in model control group (P 〈 0.05), and
