摘要
目的通过观察RNA干扰线粒体核糖体大亚基蛋白48(MRPL48)基因表达对Hela细胞线粒体能量代谢的影响,探讨MRPL48基因在代谢综合征诊断中的意义。方法将MRPL48特异性shRNA片段转入体外培养的Hela细胞,48 h后采用Western blotting方法确认shRNA的干扰效果;用JC-1染色法观察线粒体膜电位的变化。结果 MRPL48 shRNA转染细胞后,能够有效抑制Hela细胞MRPL48基因的表达;JC-1染色显示,RNA干扰组Hela细胞较对照组绿色荧光颗粒明显增加,提示线粒体膜电位下降。结论 MRPL48是维持线粒体正常功能的关键蛋白质,该蛋白在细胞代谢过程中起到了非常重要的作用,提示MRPL48可作为代谢综合征临床诊治的新靶点。
Objective To evaluate the significance of Mitochondrial Ribosomal Protein L48(MRPL48) gene in diagnosis of metabolic syndrome through observing the effects of RNA interference on mitochondrial energy metabolism.Methods Cultured Hela cells were infected with MRPL48 targeting shRNA,and the gene silencing effects verified by Western blotting after 48 h.The mitochondrial membrane potential changes were assessed using JC-1 staining.Results ShMRPL48 interfered effectively the expression of MRPL48 gene.JC-1 staining showed increased green fluorescence particles in Hela cells of RNA interference group,suggesting the decline of mitochondrial membrane potential.Conclusion MRPL48,one of the key proteins that maintain the mitochondrial functions,might play a very important role in energy metabolism,suggesting that MRPL48 can be a new target for diagnosis and treatment of metabolic syndrome.
