摘要
①目的探讨胎盘组织中水通道蛋白3(Aquaporins 3,AQP3)的表达及其与妊娠期糖尿病(Gestational diabetes mellitus,GDM)的关系。②方法选取2017年8月~2018年8月华北理工大学附属医院产科孕妇120例,其中GDM孕妇60例为GDM组,糖耐量正常孕妇60例为对照组。收集研究对象一般资料,胎盘娩出后立即取胎盘组织标本,采用Western blot法、实时荧光聚合酶链反应(RT-PCR)法分别检测两组胎盘组织中AQP3蛋白和mRNA的表达水平;分析胎盘组织中AQP3的表达与GDM的关系。③结果Western blot检测结果显示GDM组AQP3蛋白的相对表达量(0.62±0.13),对照组(0.93±0.20),AQP3蛋白在GDM组胎盘组织中的表达比对照组弱,两组比较差异具有统计学意义(P<0.05)。RT-PCR检测显示GDM组AQP3 mRNA水平(0.82±0.26),对照组(1.80±0.39),两组比较差异具有统计学意义(P<0.05)。Logistic回归分析显示AQP3蛋白表达与GDM有关,AQP3蛋白表达降低的患者发生GDM的风险是不降低患者的5.29倍(OR=5.29,95%CI:2.39~11.68)。④结论胎盘中AQP3蛋白表达与GDM有关,AQP3蛋白表达水平降低的患者发生GDM的风险增加。
Objective To explore the relationship between the expression of AQP3 in placental tissue and GDM.Methods A total of 120 cases were selected from the obstetrics department of the affiliated hospital of North China University of Science and Technology,including 60 pregnant women with GDM in GDM group and 60 pregnant women with normal glucose tolerance in control group.General data of the study subjects were collected.Placental tissue samples were taken immediately after delivery of the placenta,and the expression levels of AQP3 protein and mRNA in the placental tissues of the two groups were detected by Western blot and real-time fluorescence polymerase chain reaction(RT-PCR).The relationship between AQP3 expression and GDM in placental tissues was analyzed.Results Western blot results showed that the expression of AQP3 protein in the placental tissue of GDM group was weaker than that of the control group.The relative expression of AQP3 protein in the GDM group(0.62±0.13)was lower than that of the control group(0.93±0.20),and the difference was statistically significant(P<0.05).Rt-PCR results were consistent with Western blot results,showed that AQP3 mRNA level in GDM group(0.82±0.26)was lower than that in control group(1.8±0.39),with significantly difference(P<0.05).Logistic regression analysis showed that AQP3 protein expression was related to GDM,and the risk of GDM in patients with reduced AQP3 protein expression was 5.29 times higher than those without reduced AQP3[OR=5.29,95%CI:2.39~11.68].Conclusion The expression of AQP3 protein in placenta is related to GDM,and the risk of GDM is increased in patients with decreased AQP3 protein expression.
