五大连池小孤山火山灰细菌、古菌多样性及其同理化因子的关系 被引量:6

Correlation Between the Diversity of Tephra Bacteria and Archaea and Their Environmental Factors in Xiaogushan, Wudalianchi

摘要 对五大连池小孤山火山灰中细菌、古菌的多样性特征及其同环境因子的相关性进行了研究和探讨。小孤山火山灰的pH为7.06,总氮含量为0.01%,总碳含量为0.16%,氨氮含量为2.5μg·g^-1,硫化物含量为0.25μg·g^-1,可溶性总铁含量为3.00μg·g^-1。利用细菌和古菌的16SrRNAV3+V4区的通用引物分别扩增,以火山灰总DNA为模板,分别扩增并测定细菌的16SrRNA序列56511条,古菌测序共获得70250条序列。根据16SrRNA序列相似性97%为原核生物种的定义标准,这些序列分别定义为2015个细菌操作分类单位,和565个古菌操作分类单位,表明小孤山火山灰中细菌和古菌具有较丰富的系统发育多样性。结合环境因子分析,氨氮和硫化物与火山灰中大多数细菌和古菌的相关性呈极显著相关,推测氨氮和硫化物是影响火山灰细菌和古菌多样性的重要因素之一。 The diversity characteristics of bacteria and archaea in tephra from Xiaogushan in Wudalianchi and the correlation with environmental factors were studied and discussed. The pH of tephra in Xiaogushan was 7.06, total nitrogen content was 0.01%, total carbon content was 0.16%, ammonia nitrogen content was 2.5 μg·g^-1, sulfide content was 0.25 μg·g^-1, and soluble total iron content was 3.00 μg·g^-1. The general primers of 16S rRNA V3+V4 region of bacteria and archaea were amplified respectively, bacterial sequencing obtained 56 511 sequences, archaeal sequencing obtained 70 250 sequences. According to the 16S rRNA sequence similarity of 97% being used as the prokaryotic species declination, the bacteria could be divided into 2 015 operational taxonomic units, and archaea could be divided into 565 operational taxonomic units, indicating that there were abundant phylogenetic diversities of bacteria and archaea in tephra from Xiaogushan. Combined with environmental factor analysis, the correlation between ammonia nitrogen and sulfide and most bacteria and archaea in tephra was highly significant. It was speculated that ammonia nitrogen and sulfide were the important factors that affected the diversity of bacteria and archaea in tephra.
