摘要
为了解五大连池东焦得布山中硫氧化细菌多样性同环境因子的关系,测得9个东焦得布山火山土壤理化参数,并利用硫氧化细菌功能基因soxB作为标记物,通过设计引物采用高通量测序方法对其硫氧化细菌多样性进行测定,最后通过冗余分析(RDA分析)来解读东焦得布山中硫氧化细菌多样性同环境因子的关系。东焦得布山火山土壤理化测定结果如下:pH为7.65;ORP测定结果为49.33 mV;Fe^2+含量为0.11μg·g^-1;TOC含量为18.50%;TON含量为110.67μg·g-1,Nitrate-N含量为497.54μg·g^-1,S0含量为0.85 mg·g^-1、S2-含量为0.375 mg·g^-1、SO42-含量为541.46μg·g^-1。高通量测序结果共获得441个OTU,其主要优势菌群为慢生根瘤菌属bradyrhizobium(25.6%)、芽生绿菌属Blastochloris(12.3%)、雷尔氏菌属Ralstonia(10.3%)、贪铜菌属Cupriavidus(9.3%)、紫色硫细菌属Allochromatium(7.5%)等。这些硫氧化细菌优势菌群均属于变形菌门,通过RDA分析表明,S2-与多数菌属均呈现正相关,是影响东焦得布山硫氧化细菌群落的主要因素。
To explore the relationship between sulfur-oxidizing bacteria diversity and environmental factors in Dongjiaodebu volcano of Wudalianchi,some physical and chemical parameters were measured and the sulfur-oxidizing bacteria diversity in Dongjiaodebu volcano was conducted high-throughput analysis by using soxB as molecular biomarkers,and redundancy analysis(RDA)was used to explain the relationship.The pH of Dongjiaodebu volcano was 7.65,ORP 49.33,Fe^2+ content 13.05 mg·g^-1,TC content 18.50%,TON content 110.67μg·g^-1,Nitrate-N content 497.54μg·g^-1,S0 content 0.85 mg·g-1,S2-content 0.375 mg·g^-1,SO42-content 541.46μg·g^-1.A total of 441 OTU distributions were obtained in Dongjiaodebu volcanio,its dominant sulfur-oxidizing bacteria community mainly composed of bradyrhizobium(25.6%)、Blastochloris(12.3%)、Ralstonia(10.3%)、Cupriavidus(9.3%)、Allochromatium(7.5%).The RDA analysis showed that S2-was positively correlated with all the genus which was the main factor affecting the sulfur-oxidized bacteria community in Dongjiaodebu volcano.
