摘要
目的研究长链非编码RNA DANCR在神经胶质瘤组织中的表达水平及其对胶质瘤患者临床预后的影响。方法RT-PCR法分别检测神经胶质瘤组织样本(n=52)以及正常脑组织样本(n=23)中DANCR的表达水平,分析其与胶质瘤患者临床病理学特征之间的相关性,用Kaplan-Meier法分析患者生存率,并分析影响胶质瘤患者临床预后的因素。结果 DAN-CR在胶质瘤组织标本中的表达水平显著高于正常脑组织标本(3. 06±0. 28 vs 1. 00±0. 22,P <0. 001),随着胶质瘤病理分级的增高,DANCR的表达水平也随着增高(P=0. 013)。DANCR的表达水平与胶质瘤患者的病理分级、KPS评分以及术后复发具有相关性,而与患者的年龄、性别、肿瘤大小、肿瘤数目等无关。DANCR低表达水平组患者术后5年生存率显著高于DANCR高表达水平组。高表达水平的DANCR、高病理级别、KPS评分<80分以及术后复发均为影响胶质瘤患者临床预后的独立危险因素。结论胶质瘤组织标本中DANCR的表达水平显著升高,且高表达水平的DANCR可作为预测胶质瘤患者预后的独立危险因素。
Objective To investigate the expression level of long non-coding RNA DANCR in glioma tissue and its effect on clinical prognosis of glioma patients.Methods The expression of DANCR in human glioma tissue specimens and normal brain tissue specimens was detected by RT-PCR.Then,the correlation was analyzed between clinicopathological features and the clinical prognosis of glioma patients.After analyzing the survival rate of patients with Kaplan-Meier method,factors affecting clinical prognosis of glioma patients were analyzed at last.Results The expression level of DANCR in glioma tissue specimens was significantly higher than that of normal brain tissue specimens(3.06±0.28 vs1.00±0.22,P<0.001),and the expression level of DANCR was also increased with the increase of pathological grade of glioma(P=0.013).The expression level of DANCR was correlated with pathological grading,KPS score and postoperative recurrence in patients with glioma,but had nothing to do with age,sex,tumor size and number of tumors.The 5-year survival rate of patients with low expression level of DANCR was significantly higher than that of high expression level.High expression levels of DANCR,high pathological grade,KPS score<80 and postoperative recurrence were independent risk factors for clinical prognosis in patients with glioma.Conclusion The expression level of DANCR in glioma tissue specimen is significantly up-regulated.High expression level of DANCR could be used as an independent risk factor in predicting the prognosis of patients.
