微小RNA-129在神经胶质瘤组织中的表达水平和临床价值

摘要 目的分析微小RNA-129非编码RNA在神经胶质瘤组织中的表达水平及其临床意义。方法选取62例神经胶质瘤患者为观察组,62例同期健康志愿者为对照组。提取观察组组织和对照组总RNA、逆转录为cDNA,实时荧光定量RT-PCR法分析胶质瘤组织样本及正常脑组织样本微小RNA-129的表达水平,并分析其与临床病理指标和预后的关系。结果微小RNA-129在胶质瘤组织的表达水平低于正常脑组织标本[(2.75±0.28)vs(1.00±0.13)],且WHO分级Ⅲ~Ⅳ级患者低于Ⅰ~Ⅱ级患者[(2.41±0.22)vs(3.71±0.40)],差异均有统计学意义(P<0.05)。设置微小RNA-129在神经胶质瘤的表达平均数3.08为界,微小RNA-129低表达组的患者预后与高表达组比较,患者术后5 a生存期大大缩短,差异有统计学意义(P<0.05)。WHO分级、KPS评分、微小RNA-129是影响神经胶质瘤患者预后的危险因素。多因素COX比例分析显示,微小RNA-129低表达水平、高WHO分级及KPS评分≤80分是影响神经胶质瘤患者预后的独立危险因素。结论微小RNA在神经胶质瘤组织表达水平降低,并与临床病理特征和预后密切相关。 Objective To study the expression of microRNA-129 non coding RNA in gliomas and its influence on the clinical prognosis of gliomas.Methods 62 cases of gliomas and 62 healthy volunteers were selected as the control group.The expression of microRNA-129 in glioma tissue and normal brain tissue was analyzed by Real-time Fluorescence Quantitative RT-PCR,and the relationship between microRNA-129 and clinicopathological indexes and prognosis was analyzed.Results The expression level of microRNA-129 in gliomas was significantly lower than that in normal brain tissues(2.75±0.28)vs(1.00±0.13),and the level of microRNA-129 in WHO gradeⅢ-Ⅳpatients was lower than that inⅠ-Ⅱpatients(2.41±0.22)vs(3.71±0.40),there were significant difference(P<0.05).The mean expression of microRNA-129 in glioma was set as 3.08,the prognosis of patients with low expression of microRNA-129 was significantly worse than that of patients with high expression,and the 5-year survival period of patients after operation was significantly shorter,the difference was statistically significant(P<0.05).Univariate analysis showed that WHO grade,KPS score and microRNA-129 were risk factors for prognosis of patients with glioma.Multivariate COX analysis showed that low expression level of microRNA-129,high WHO grade and KPS score≤80 were independent risk factors for the prognosis of gliomas.Conclusion The expression level of microRNA-129 in gliomas is significantly decreased,which is closely related to clinicopathological features and prognosis.
