琐琐葡萄多糖对PM2.5致大鼠肺损伤保护作用研究 被引量:6

摘要 目的研究琐琐葡萄多糖(Vitis vinifer L.VTP)对PM2.5所致大鼠肺损伤的保护作用。方法 48只SPF级Wistar大鼠随机分为对照组、枸杞多糖(Lycium Barbarum Polysaccharide,LBP)组(500mg/kg体质量)、染尘组及VTP高、中、低剂量组(500、250、125mg/kg体质量),每组8只。对照组、染尘组蒸馏水灌胃,其余各组多糖灌胃30d;第31天无创气管滴注PM2.5混悬液(15mg/kg体质量),对照组滴注生理盐水。检测血清中乳酸脱氢酶(LDH)、碱性磷酸酶(ALP),肺灌洗液(bronchoalveolar lavage fluid,BALF)中ALP、LDH、肿瘤坏死因子(TNF-α),肺组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA),肺湿干重比(W/D)与肺、脾、胸腺指数,以及病理观察。结果染尘组大鼠血清中ALP、LDH升高,VTP各剂量组降低(P<0.05);染尘组BALF中ALP、LDH升高,VTP各剂量组ALP降低,VTP中、高剂量组LDH降低(P<0.05)。染尘组BALF中TNF-α升高,VTP各剂量组TNF-α水平降低(P<0.05)。染尘组SOD活力降低、MDA含量升高,VTP高剂量组SOD活力升高,各剂量组MDA含量下降(P<0.05)。染尘组W/D升高,VTP中、高剂量组W/D降低(P<0.05)。染尘组脾指数降低,VTP低、中剂量组脾指数升高(P<0.05);胸腺指数各组之间差异无统计学意义染尘组肺指数升高,VTP各剂量组肺指数下降(P<0.05)。结论琐琐葡萄多糖对PM2.5所致大鼠肺损伤有一定保护作用。 Objective To observe the effect of Vitis vinifer L.(VTP)on lung injury induced by PM2.5in rats.Methods 48 Wistar rats were randomly assigned into the NS control,PM2.5,PM2.5+LBP and PM2.5+VTP groups(500,250,125mg/kg),8rats in each group.Rats in control and PM2.5groups were given water,rats in remaining groups were given polysaccharide for 30 days before the induction of tracheal instillation.Rats were given PM2.5suspension(15mg/kg,tracheal instillation)to induce lung injury.The ALP,LDH,TNF-αin BALF and SOD,MDA in tissue,W/D of lung and organ index of tissue were measured.Results The ALP,LDH in serum and BALF of PM2.5group were higher than control group,ALP,LDH in serum of medium and high dose VTP groups were lower than PM2.5group;ALP in each VTP groups were lower than PM2.5group;LDH in medium and high dose VTP groups were lower than PM2.5group(P〈0.05).TNF-αin BALF of PM2.5group was higher than control group and each VTP groups was lower than PM2.5group.Compare to control group,SOD was decreased and MDA wasincreased in PM2.5group;compare to PM2.5group,SOD was increased and MDA was decreased in each VTP groups(P〈0.05).W/D of PM2.5group was higher than control group,while medium and high dose VTP groups were lower than PM2.5group(P〈0.05).Index of spleen of PM2.5group was lower than control group,each VTP groups was increased(P〈0.05).There was no statistically significant difference among the groups of index of thymus.Index of lung of PM2.5group was higher than control goup,while medium and high dose VTP groups were decreased(P〈0.05).Conclusion The results indicated that VTP has a protective effect on PM2.5induced lung injury in rats.
