两种止咳药物对大气PM2.5致大鼠急性肺损伤的防治作用研究 被引量:2

摘要 目的研究治咳川贝枇杷露、蜜炼川贝枇杷膏对PM2.5致大鼠急性肺损伤(ALI)的预防和治疗作用。方法雄性SD大鼠随机分为正常对照组,枇杷露对照组,枇杷膏对照组,PM2.5模型组,枇杷露预防和治疗低、中、高剂量组,枇杷膏预防和治疗低、中、高剂量组,每组6只;除对照组外,其余各组分别在给药前或给药后以PM2.5悬浊液气管滴注方法建立ALI模型,观察各组大鼠的一般状态、肺脏形态和肺组织病理学改变,收集肺泡灌洗液(BALF),测定肺重比、生化指标酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、乳酸脱氢酶(LDH)、白蛋白(ALB)、总蛋白(TP)及炎症因子白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)等的含量。结果与正常对照组比较,枇杷露和枇杷膏对照组对大鼠的体表状态和肺脏形态无影响,3个对照组的肺重比、ACP、AKP、LDH、ALB、TP、IL-6、TNF-α等含量的差异无统计学意义,PM2.5模型组大鼠出现明显呼吸哮鸣音,肺内见颗粒物沉积,肺组织切片呈现急性肺损伤改变,BALF中生化指标及炎症因子浓度均显著升高(P<0.01);与PM2.5模型组比较,枇杷露、枇杷膏的预防组和治疗组中大鼠的体表状态、肺脏形态和病理改变均得到显著改善,哮鸣音减弱,颗粒物沉积变少,BALF中上述指标值均明显降低(P<0.01、P<0.05)。结论治咳川贝枇杷露和蜜炼川贝枇杷膏对PM2.5所致ALI具有明显的预防和治疗作用,可缓解肺损伤症状,减少肺内生化指标和炎症因子的分泌,保护肺部组织。 Objective To determine the prevention and therapy of PM2.5-induced acute lung injury with Zhike Chuanbei Pipa Lu and Milian Chuanbei Pipa Gao in rats.Methods Male SD rats were randomly divided into a normal control group,a Pipa Lu control group,a Pipa Gao control group,a PM2.5 exposure group,3 Pipa Lu prophylaxis and treatment groups,and 3 Pipa Gao prophylaxis and treatment groups,6 rats in each group.Acute lung injury rat model was estabilished by intra-tracheal instillation with suspension of PM2.5.The physical signs,weight of lung,and pathological changes in the lung tissue were observed.The content of ACP,AKP,LDH,ALB,TP,IL-6 and TNF-α was determined.Results Compared with the normal control group,Pipa Lu and Pipa Gao control groups had no effect on the physical signs and lung morphology of the rats.There was no significant difference in the lung index,ACP,AKP,LDH,ALB,TP,IL-6,and TNF-α of the three control groups.The PM2.5 exposure group had obvious respiratory wheezing sounds and intrapulmonary particulate matter deposition.The lung tissue slices showed acute lung injury changes and the concentrations of biochemical markers and inflammatory factors in BALF were increased significantly(P<0.01).Compared with the PM2.5 exposure group,the lung morphology and pathological changes of Pipa Lu and Pipa Gao prophylaxis and treatment groups were all significantly improved,the deposition of particulate matter was reduced,the wheezing sound weakened,and the above indexes in BALF were significantly decreased(P<0.05 or P<0.01).Conclusion Pangaoshou Zhike Chuanbei Pipa Lu and Milian Chuanbei Pipa Gao may obviously prevent and treat PM2.5 induced respiratory tract injury,relieve the secretion of inflammatory factors and various cell contents,and reduces lung injury and protect lung tissue.
