天祝白牦牛KAP1.1基因的克隆及生物信息学分析 被引量:2

摘要 以牦牛的角蛋白关联蛋白1.1(KAP1.1)基因为研究对象,根据GenBank收录的黄牛KAP1.1基因的mRNA序列(登录号:NM_001105369.1)设计特异性引物,通过反转录PCR、PCR以及克隆方法首次获得牦牛的KAP1.1完整CDS区,将所得序列提交到NCBI,登录号为KJ095199,并对其进行生物信息学分析。结果表明,牦牛KAP1.1基因的CDS区全长为501bp,共编码166个氨基酸;编码的蛋白质属于亲水性蛋白质。二级结构含有α螺旋区、延伸链、β转角和无规卷曲4种。氨基酸序列与黄牛的同源性最高,具有High sulphur keratin-associated protein家族蛋白的完整结构域。 Taking Keratin associated protein 1.1(KAP1.1) gene of yak as the research object,a specific primer wasdesigned according to the m RNA sequence of cattle KAP1.1 gene(Accession No.NM_001105369.1).Yak's complete CDS sequences of KAP1.1 were obtained by RT-PCR,PCR and cloning,the sequences were submitted to NCBI(Accession No.KJ095199) and analyzed by bioinformatics.The results showed that,the length of CDS sequences in yak KAP1.1 gene was 501 bp encoding 166 amino acids which belonged to hydrophilic protein.The secondary structures were mainly composed of the alpha helix,extended strand,Beta turn and random coil.The amino acid sequence had the highest homology with cattle,and it had the complete family proteinsdomain of High sulphur keratin-associated protein.
