摘要
本研究以768只中国美利奴羊(新疆军垦型)为实验材料,采用测序和PCR技术分析KAP1.1(B2A)基因单核苷酸多态性。测序表明中国美利奴羊(新疆军垦型)KAP1.1基因存在30 bp的插入/缺失,12%PAGE电泳证实KAP1.1基因存在6种基因型:(341 bp)AA型、(311 bp)BB型、(281 bp)CC型、(341 bp/311 bp)AB型、(341 bp/281 bp)AC型和(311 bp/281 bp)BC型,基因型频率依次为0.0208、0.5534、0.0260、0.1589、0.0456和0.1953。检测到A、B和C3个等位基因,基因频率依次为0.1230、0.7305和0.1465。研究结果将为开展优质细毛羊毛性状功能基因的研究提供基础。
Sequencing and SNP snalysis was used to identify sequence variation in KAPI. 1 (B2A)gene. 12 % PAGE electrophoresis was used to identify length polymorphism in the KAPI. 1 gene. The allelic variation was confirmed by DNA sequencing. The results showed three KAP1.1 alleles were identified which varied in length due to the insertion or deletion of 30 nucleotldes. There were seven genotypes of KAP1.1 gene,which were(341 bp)AA genotype,(311 bp)BB genotype,(281 bp)CC genotype,(341 bp/311 bp)AB genotype,(341 bp/ 281 bp) AC genotype and (311 bp/281 bp ) BC genotype, respectively. Genotypic frequency were by turns 0. 0208,0. 5534,0. 0260,0.1589, 0. 0456 and 0. 1953. A,B and C allele frequency were 0. 1230,0. 7305 and 0. 1465. This study would provide basis for finding functional gene of wool trait.
