利用自主发光结核分枝杆菌快速连续检测抗结核药物的细胞内活性 被引量:5

摘要 目的:建立无筛选标记的自主发光结核分枝杆菌感染巨噬细胞模型并利用该模型直接快速连续检测抗结核药物的细胞内活性。方法:将小鼠巨噬细胞株(Raw264.7)接入96孔板中贴壁过夜处理,然后利用UAlRv菌感染4 h后进行清洗,加入抗结核药物后进行培养。分别在感染后的第0~7天对同一样品进行发光值的检测,直接通过发光值的变化进行药效的判断。结果:成功构建UAlRv感染小鼠巨噬细胞的模型,并利用该模型成功对利福平(RIF),异烟肼(INH),莫西沙星(MFX)和利奈唑胺(LZD)等抗结核药物进行细胞内抗结核作用评价。UAlRv体外检测药物活性的最小抑菌浓度(MIC,μg/mL)分别为RIF 0.125,INH 0.06,MFX 0.5和LZD 2.0,与之前报道过的利用带抗性标记的自主发光结核杆菌体外检测药物活性一致。在UAlRv感染巨噬细胞模型中药物的MIC(μg/mL)分别为RIF 0.125,INH 0.015,MFX 0.5和LZD 4.0,这与报道过的利用普通H37Rv结核菌感染巨噬细胞模型检测的结果相一致。结论:利用UAlRv感染巨噬细胞模型可快速、经济、连续地检测抗结核药物的细胞内活性,并可能用于高通量筛选/评价抗结核药物。
