摘要
以1 400份大豆品种(系)资源为材料,利用SDS-PAGE,分析贮藏蛋白11S和7S组分及其亚基含量与亚基缺失情况,并采用0.2%,0.4%EMS分别处理南农493-1和南农99C-6两个品种种子,利用60Coγ射线处理76个品种(系)种子,处理之后及时播种,单株收获,SDS-PAGE检测M2代和M3代种子贮藏蛋白亚基.结果表明,1 400份参试品种(系)的11S/7S值范围0.44~3.64,平均值1.68±0.37,并鉴定出34份亚基含量特异材料,其中包括α′和α亚基含量低、β亚基含量低、A3B4亚基缺失、A5A4B3亚基缺失和A1aB1b亚基缺失种质.理化诱变结果表明,化学诱变率约为2.92%,物理诱变率约为5.71%,对大豆蛋白的诱变效果,0.4%EMS处理优于0.2%EMS处理,而60Coγ射线慢照射处理优于化学诱变处理,并从M3代种子中筛选出了211粒亚基变异种质.
The contents of 7S and 11S globulins and their respective subunits, and the 11S/7S ratios in seeds of 1 400 soybean varieties were analyzed using SDS-PAGE, and some seeds from cultivars Nannong 493-1 and Nannong 99C-6 were treated by 0.2%, 0.4%EMS respectively, 60^Coγ-ray irradiated some seeds of 76 soybean cultivars. The average and variation range of the 11S/7S ratios of 1400 soybean varieties were 1.68±0.37 and 0.44-3.64, and 34 variation accessions of subunits content were identified, including low level of α′, α-subunit, low level of β-subunit, absence of A3B4-subunit, absence of A5A4B3-subunit and A1aB1b-SUbunit. The results of induced variation germplasm of storage proteins by physical and chemical method indicated: the chemical inducement rate is 2.92%, while the physical inducement rate is 5.71%, furthermore,60^Coγ-ray irradiation treatment 〉 0.4%EMS treatment 〉 0.2% EMS treatment, and 211 seeds of subunits variation were detected in the M3 seed.
