栽培大豆蛋白亚基11S/7S组成及过敏蛋白缺失分析 被引量:32

摘要 以175份中国大豆品种资源为材料 ,利用SDS PAGE电泳和Western杂交技术 ,分析蛋白亚基 1 1S和 7S的含量 ,以及GlymBd 2 8K和GlymBd 30K两种过敏蛋白的缺失情况。结果表明 ,参试品种的 1 1S和 7S的含量呈极显著负相关(r=-0 95) ,1 1S 7S比值范围为 0 77~ 4 67,并鉴定出 1份自然缺失 β亚基的材料。在 1 75个品种中没有检测到缺失30K过敏蛋白的材料 ,但发现 77份缺失 2 8K过敏蛋白的品种 ,占被检测品种的 44 0 %。蛋白质高于 43 0 %的品种和蛋白质含量低于 43 0 %的品种中 2 8K过敏蛋白的缺失比例分别为 56 0 %和 2 8 0 % ;地方品种 ( 1 2 6份 )和选育品种 ( 49份 )中缺失 2 8K过敏蛋白比例分别为 48 4 %和 32 7% ;在北方春大豆、黄淮夏大豆和南方大豆中 ,2 8K过敏蛋白缺失的品种分别占 1 1 7%、60 7%和 60 9%。而这 3个栽培区的大豆品种中蛋白质含量高于 43 0 %的品种比例分别为 1 0 0 %、57 1 %和 88 5%。相关分析表明 ,2 8K过敏蛋白的缺失与蛋白质含量正相关 (r=0 1 4 )。 Protein subunit contents of 175 Chinese soybean cultivars were analyzed by using SDS PAGE, the ratio of 11S/7S arranged from 0 77 to 4 67, and a negative correlation between the contents of 11S and 7S was found. A mutant cultivar lacking the β subunit of 7S was found at the same time. Western blotting was used to identify the allergenic protein Gly m Bd 28K and Gly m Bd 30K of the 175 cultivars. None materials were found lacking the Gly m Bd 30K allergen, but 77 were found lacking the Gly m Bd 28K allergenic protein, accounted for 44 0% of the tested materials. In the materials with protein content higher or lower than 43%, 56 0% and 28 0% cultivars were lacking in the Gly m Bd 28K allergen, respectively. There were 48 4% and 32 7% cultivars lacking Gly m Bd 28K allergen in the local varieties and breeding lines. Cultivars lacking Gly m Bd 28K allergen accounted for 11 7%, 60 7% and 60 9% in north spring soybean, Huanghuai summer soybean, and south China soybean, respectively, where these cultivars with protein content more than 43% were 10 0%, 57 1% and 88 5%. The result showed that the lacking of Gly m Bd 28K allergen was correlated to the protein content ( r =0 14).
