Molecularly Imprinted On-line Solid-phase Extraction Combined with Flow Injection Chemiluminescence for Determination of Chlortetracycline

摘要 基于盐酸金霉素在酸性介质中能极大的增敏Ce(Ⅳ)和Rh6G的化学发光,建立了结合分子印迹固相萃取的高灵敏度的化学发光测定盐酸金霉素的方法。用于盐酸金霉素结构相似的四环素作为虚拟模板合成了对盐酸金霉素有特异吸附的分子印迹聚合物,将填充有分子印迹聚合物的聚四氟乙烯管作为固相萃取柱,连接在八通阀上对盐酸金霉素进行萃取和预富集,用洗脱液(V(甲醇)∶V(0.01mol/LHNO3)=1∶4)在线洗脱吸附在固相萃取柱上的盐酸金霉素,对盐酸金霉素在检测池中与Ce(Ⅳ)和Rh6G在酸性条件下发生的化学发光强度进行测定。结果表明,盐酸金霉素的线性范围为2×10^-8-1×10^-6g/mL,方法的检出限为4×10^-9g/mL(3σ),7次平行测定2×10^-7g/mL的盐酸金霉素溶液的化学发光强度相对标准偏差为0.65%。用于鸡肝脏中盐酸金霉素含量测定,结果令人满意。 Chlortetracycline hydrochloride can enhane the chemiluminescence of Ce (Ⅳ) and Rh6G in acidic medium. So novel molecularly imprinted polymer solid-phase extraction(MISPE) method combined with flowinjection chemiluminescence for determining chlortetracycline hydrochloride was developed. The MIP was synthesized by using tetracycline as the imprinting molecule which has a structure similar to that of chlortetracycline. A polytetrafluoroe-thylene (PTFE) tube packed with the particles of MIP was employed as an MISPE micro-column and was connected to the sampling loop of the eight-way injection valve for the preconcentration and extraction of chlortetracycline hydrochloride. The eluent was used as the carrier for extracting the adsorbed chlortetracycline hydrochloride to react with a mixture of cerium ( Ⅳ ) and rhodamine6G in the flow cell to produce strong CL. Intensity of CL is linearly changed with the concentration of chlortetracycline hydrochloride from 2 × 10^-8 g/mL to 1 × 10 ^-6 g/mL and the limit detection is 4 × 10^-9 g/mL(3σ). This method has been successfully applied to the determination of chlortetracycline hydrochloride in liver of chicken sample.
