摘要
通过BSA法,对甘蓝型油菜核不育材料S45A的育性基因进行分子标记,在980个随机引物中,找到了与可育基因连锁的2个RAPD标记UBC158-600、UBC187-600,其遗传图距分别为9.564cM(LOD,21.495)、17.700cM(LOD,13.244)。
Bulked segregant analysis was employed to identify random amplified polymorphic DNA(RAPD) marks linked to the Ms gene used in the recessive genetic male sterility of rapeseed (B. napus). A total of 980 arbitrary 10- mer oligonucleotide primers were screened on the DNA of fertile and. sterile bulks. Two primers, C158 and C187, produced repeatable polymorphism between the paired bulks. DNA from individual plants of each bulk was then used as a template for amplification with these two primers. DNA polymorphisms generated by two primers were found to be tightly linked to the Ms gene. Their genetic distance to the ms gene was 9. 564 cM (LOD 21. 495) and 17. 700 cM (LOD 13.244) respectively.
