摘要
对甘蓝型油菜“中油杂 2号”杂交种及其父母本的酯酶同工酶分析结果显示,父本和杂种F1 比母本多一条迁移率(Rf)为 0. 8的条带,该特征条带可用来进行杂种纯度鉴定。在“中油杂 2号”的父母本间筛选了 20个AFLP引物组合,发现E33M59不仅扩增产物多态性好,而且有 3条母本特异带和 7条父本特异带在杂种F1中出现,因此该引物组合被用来进行“中油杂 2号”种子纯度的鉴定。对 5个“中油杂 2号”样品进行纯度鉴定的结果表明,酯酶同工酶和AFLP标记鉴定与田间种植鉴定的结果相吻合。
The results of the analysis on esterase isozyme bands of Brassica napus hybrid “Zhongyouza No.2”and its parents indicated that one specific band with a relative mobility (Rf) of 0.8 presented in hybrid and male parent, but absent from female parent. So this specific band can be used to identification of hybrid. Among 20 AFLP primer combinations, E33M59 had the most abundant polymorphism between the sterile line and the restoration line. 7 specific bands from the male parent and 3 specific bands from the female parent presented in hybridity, so E33M59 can be used to identification of hybridity. The results of the test on 5 samples of “Zhongyouza No.2” showed seed purity identified by esterase isozyme and AFLP markers tallied with the purity identified by planting in field.
