摘要
利用共显性分子标记技术SSR、显性形态标记性状花叶及显性生化标记种子芥酸含量鉴定甘蓝型油菜自交不亲和系SI-1300与恢复系花叶恢杂种F1种子纯度。结果表明,3种方法鉴定结果基本一致,F1杂种种子纯度达到91%以上;不同SSR引物鉴定结果完全相同,一对SSR引物即可用于鉴定F1杂种纯度。SSR扩增产物具有很高的多态性和严格的保守性,不仅可以用于油菜杂种纯度鉴定,亦可以用于不同品种(组合)真实性鉴别。研究结果还表明,自然条件下放养蜜蜂用于自交不亲和系杂种制种是一种稳妥、切实可行的方法。
To determinate seed purity of F_1 hybrid produced by self-incompatible line SI-1300 and self-compatible variety Hua Yehui in Brassica napus, microsatellite (SSR) and two marker traits, full-lobe leaf and seed erucic acid concentration, were used in this study. The result obtained from SSR marker was essentially consistent with those from the two traits, and the F_1 seed purity was over 91%. Only one primer pair of SSR was enough for hybridity identification. The result also indicated that honeybee-assisted pollination was helpful for hybrid production of self-incompatiblity in Brassica napus.
