摘要
本试验采用响应面设计法优化光合细菌培养基。结果表明:培养基成分中醋酸钠和蛋白胨对于光合细菌的生长影响最为显著,最优培养基配方为:醋酸钠1.145 g/L、蛋白胨0.055 g/L、碳酸氢钠0.6 g/L、硫代硫酸钠0.4 g/L、氯化钠0.3 g/L、硫酸镁0.1 g/L、磷酸二氢钾0.05 g/L。在此条件下,光合细菌生长最为良好,经过5 d培养以后,培养液OD600可以达到0.5以上。
Response surface methodology was used to optimize the culture medium for Photosynthetic bacteria(PSB). The result showed that sodium acetate and peptone has the most significant influence on PSB cultures. The optimized medium for PSB culture was as follows: sodium acetate 1. 145 g/L, Peptone 0. 055 g/L,NaHCO3 0.6 g/L,Na2S2O3 0.4 g/L, NaCl 0.3 g/L, MgSO4 0.1 g/L, KH2PO4 0. 05 g/L. Under this condition,the OD600 of the broth reached a higher level of 0.5 after 5 day' s culture.
