摘要
用响应面法对解淀粉芽孢杆菌发酵生产褐藻胶裂解酶的培养条件进行了优化。用单因数实验逼近各因素的最大响应区域,利用Plackett-Burman试验设计筛选出影响酶活的3个主要因素,即装液量、pH和蛋白胨浓度。并在此基础上利用Box-Behnken试验设计及响应面方法进行回归分析。通过求解回归方程得到优化发酵条件:当装液量65.28 mL,pH 7.35和蛋白胨浓度0.44%时,液体发酵液中褐藻胶裂解酶酶活达到理论最大值6.48 U/mL。经3批培养验证,预测值与验证试验平均值接近。
Using response surface methodology to optimize the fermentation conditions of Bacillus amyloliquefaciens producing alginate lyase,the detail steps are as follows: at first,using single-factor method to approach maximum response regions of each fermentation conditions,and using Plackett-Burman method to screen three main influencing factors for enzyme activity(peptone concentration,pH and liquid volume).Then,using Box-Behnken method and response surface method to make regression analysis,and the optimized fermentation conditions were obtained as follows: when liquid volume was 65.28 mL,pH was 7.35 and peptone concentration was 0.44%,the enzyme activity of alginate lyase in fermentation liquid reached theoretical maximum value 6.48 U/mL.After three batches of cultivation,the maximum predicted value was consistent with mean value of verification test.
