摘要
目的:重组人溶菌酶(rhLYZ)滴眼液经家兔干眼模型的药效学实验后,测定分析兔眼房水中重组人溶菌酶的含量,综合判断rhLYZ进入房水的可能性。方法:通过SDS-聚丙烯酰胺凝胶电泳法(SDSPAGE)、免疫印迹法(Western blotting)和反相高效液相色谱法(RP-HPLC),分别对兔眼房水中的rhLYZ进行定性分析及定量检测。结果:经3种方法对测定结果的比对及综合分析,用药组兔眼房水中,准确定性和定量检测到rhLYZ的样本仅有2个(n=18)。结论:经滴眼,在干眼的治疗过程中,初步判断rh LYZ基本在眼表发挥作用,通过眼表进入房水的可能性较低,或进入房水的量甚微,在药物代谢实验中需采用更为灵敏的方法进行检测分析。
Objective:To measure the content of recombinant human lysozyme in rabbit aqueous humor after pharmacodynamic test of recombinant human lysozyme(rhLYZ)eye drops on rabbit dry eye model,and comprehensively judge the possibility of rhLYZ entering aqueous humor.Methods:SDS-PAGE,Western blotting and reversed phase high performance liquid chromatography(RP-HPLC)were used to conduct qualitative analysis and quantitative detection of rhLYZ in aqueous humor of rabbits.Results:By comparing and analyzing the results of the three methods,only two samples(n=18)were accurately detected to contain rhLYZ in aqueous humor of rabbits in the model group.Conclusion:rhLYZ functions on the ocular surface during the treatment of dry eye.The possibility for it to enter aqueous humor through the ocular surface is very low,or the amount entering aqueous humor is very small.A more sensitive method should be applied in the drug metabolism test.
