对于本科实验教学中蛋清溶菌酶分离纯化实验的改进

摘要 蛋白质的分离纯化技术是生物化学的重要基础,在本科实验教学中占有重要地位。本科教学实验中的溶菌酶分离纯化实验是有助于学生了解并掌握蛋白质的分离纯化技术的经典实验,但在实际操作中有所欠缺。为了获得更好地实验结果与教学效果,本文对实验做了一定改进,利用离子交换层析、硫酸铵沉淀、分子筛层析等方法从鸡蛋清中获取溶菌酶,用SDS-PAGE电泳进行鉴定,并对溶菌酶进行了结晶、定性和定量上测定溶菌酶活力,尽量涉及了蛋白质实验的多个领域,且重复性好,易于学生操作,可更加有利于学生对蛋白质性质的理解乃至对之前所学实验的理解,培养学生的独立科研能力。 The separation and purification technology of proteins is an important foundation of biochemistry and holds an important position in undergraduate experimental teaching.The Lysozyme separation and purification experiment in undergraduate teaching experiment is a classic experiment that helps students understand and master the protein separation and purification technology,but it is lacking in practical operation.In order to obtain better experimental results and teaching effects,this paper has made some improvements in the experiment.Lysozyme is obtained from egg white by means of ion exchange chromatography,ammonium sulfate precipitation,molecular sieve chromatography and other methods,and is identified by SDS-PAGE electrophoresis.Lysozyme is crystallized,qualitatively and quantitatively determined Lysozyme activity.It involves many fields of protein experiment as far as possible,has good repeatability,and is easy for students to operate,It can be more conducive to students'understanding of protein properties and even their understanding of previous experiments,and cultivate their independent scientific research ability.
