高效液相色谱法测定配制酒中黄色素的方法学

The Methodology of the Determination of Yellow Pigment in Liquor Preparation by High Performance Liquid Chromatography

摘要 建立高效液相色谱法同时测定配制酒中栀子黄、柠檬黄、日落黄的分析测定方法。样品经加热除去乙醇后,用水定容后经微孔滤膜(0.45μm)过滤。以甲醇和0.02 mol/L乙酸铵溶液为流动相,利用高效液相色谱分析,外标法定量。结果表明,藏花素、藏花酸、柠檬黄、日落黄在1.0~20.0μg/mL范围内线性良好,相关系数r≥0.998。检出限按公式C_L=3S_b/b计算,得出藏花素2.837 mg/kg、藏花酸0.614 mg/kg、柠檬黄0.117 mg/kg、日落黄0.105 mg/kg,满足定量分析的要求。添加不同浓度的黄色素(添加量0.5~9.0 mg/kg),加标回收率在94.0%~98.5%之间, RSD<5%。该方法简单、快速,准确度高,精密度好,满足定量分析要求。 A method for the determination of gardenia yellow,lemon yellow and sunset yellow in assembled alcoholic drinks by high performance liquid chromatographic(HPLC)was established.After the sample was heated to remove the ethanol,then the extracting solution was filtered by syringe filter(0.45μm),measured by HPLC with methanol and 0.02 mol/L ammonium acetate solution as mobile phase,and external standard method was used for quantification.Results showed that the method showed a good linearity ranged from 1.0μg/mL to 20.0μg/mL of crocin,crocetin,lemon yellow and sunset yellow concentration with correlation coefficients above 0.998.The detection limit was calculated according to the formula CL=3 Sb/b,and that crocin of 2.837 mg/kg,crocetin of 0.614 mg/kg,citric yellow of 0.117 mg/kg,and sunset yellow of 0.105 mg/kg,meet the requirements of quantitative analysis.Different concentrations of yellow pigment(addition amount 0.5-9.0 mg/kg)were added,the recoveries were between 94.0%and 98.5%,and RSD was less than 5%.The method was simple,fast,accurate and accurate,and it satisfies the requirement of quantitative analysis.
