摘要
目的:探讨Ⅱ型糖尿病大鼠模型心肌细胞细胞凋亡及其调控基因表达的变化,为进一步研究NIDDM的发病机制提供实验资料。方法:建立NIDDM大鼠模型,TUNEL法观察凋亡的心肌细胞。抽提心肌组织DNA进行琼脂糖凝胶电泳。免疫组织化学方法观察心肌细胞内Bax、Bcl-2、c-fos、c-jun蛋白染色强度。结果:TUNEL法染色可见核呈棕色反应的阳性凋亡细胞,NIDDM组与对照组比较,凋亡率有高度显著差异(P<0.01)。琼脂糖凝胶电泳可见DNA梯带。NIDDM组较对照组Bax蛋白阳性平均光密度值(OD值)显著增高(P<0.01);对照组较NIDDM组Bcl-2/Bax光密度比值增高(P<0.05);NIDDM组较对照组c-fos、c-jun表达显著增高(P<0.01)。结论:实验性Ⅱ型糖尿病大鼠部分心肌细胞显示凋亡;细胞凋亡与Bax、c-fos、c-jun表达增强及Bcl-2/Bax比值降低有关。
Objective:To study myocardial apoptosis in myocardium in rats in vivo and to observe the changes of associated genes expression. Methods:Diabetes was induced by a single iv. injection of streptozotocin. TUNEL method was used for detection of apoptosis. Expression of Bcl-2/Bax,c-fos,c-jun protein were assayed by immunohistochemical technique. Image analysis was used to analyze the expressive results, extracted DNA was used to identity DNA ladder by ways of agarose gel electrophoresis. Results:TUNEL staining showed apoptosis rate of NIDDM group was obviously higher than that of control group (F<0.01). Clear DNA ladder could be found in NIDDM group. Image analysis showed the OD values of Bcl-2/Bax,c-fos and c-jun portein increased significantly compared with control group. Conclusion:To sum up,apoptosis exist in part of myocardium of NIDDM rats and it is associated with the increase of Bax,c-fos, c-jun and the decrease of Bcl-2/Bax.
