鸡腿菇产纤溶酶液体发酵体系优化

Optimization of liquid fermentation system for production of fibrinolytic enzyme from Coprinus comatus

摘要 该研究采用液态发酵法发酵鸡腿菇,鸡腿菇纤溶酶活力为评价指标,通过单因素试验、Plackett-Burman试验及Box-Behnken试验对其培养基组成和培养条件进行优化。结果表明,最佳培养基的组成为果糖2.9%,豆饼粉3.6%,KH2PO_(4)0.25%,MgSO_(4)0.2%。最佳培养条件为装液量50 mL/250 mL,接种量10%,发酵温度24℃,发酵时间6 d,摇床转速160 r/min。在此优化培养基及培养条件下,纤溶酶活力为136.89 U/mL,比优化前提高了4.28倍。 Coprinus comatus was fermented with liquid fermentation method,using fibrinolytic enzyme activity as the evaluation index,the medium composition and culture conditions were optimized by single factor experiments,Plackett-Burman experiments and Box-Behnken experiments.The results showed that the optimal medium composition was fructose 2.9%,soybean cake 3.6%,KH2PO_(4)0.25%,MgSO_(4)0.2%.The optimal culture conditions were liquid volume 50 ml/250 ml,inoculum 10%,fermentation temperature 24℃,time 6 d,and shaking speed 160 r/min.Under the optimized medium and culture conditions,the fibrinolytic enzyme activity was 136.89 U/ml,which was 4.28 times higher than that before optimization.
