摘要
在单因素试验的基础上,采用Plackett-burman试验、最陡爬坡试验和Box-behnken试验设计优化超声波辅助复合酶法提取通荞3号多糖工艺进行优化,测定多糖含量采用苯酚-硫酸法。结果表明,最优提取工艺为中性蛋白酶(50000.0 U/g)添加量0.9000%,纤维素酶(≥15000.0 U/g)添加量0.0450%,果胶酶(≥50.0 U/g)添加量0.0324%,料液比1∶20(g∶mL),pH值7.0,提取温度60℃,超声功率424 W,在此条件下多糖得率为7.31%。并对通荞3号多糖体外抗氧化能力做了相关研究,结果表明,多糖质量浓度为1.0 g/mL时清除DPPH自由基、ABTS+自由基的能力分别是62.31%和88.78%,FRAP为76.76%,由此可以为通荞3号的开发利用提供理论依据。
Based on the single factor test,the Plackett-burman test,the steepest ascent test,and the Box-behnken test design were designed to optimize the extraction process of Tongqiao No.3 polysaccharide by ultrasonic-assisted compound enzyme method.The content of polysaccharides was determined by the phenol-sulfuric acid method.The results showed that the optimal extraction process were 0.9000%neutral protease(50000.0 U/g),0.0450%cellulase(≥15000.0 U/g),0.0324%pectinase(≥50.0 U/g),solid-liquid ratio 1∶20(g∶mL),pH 7.0,extraction temperature at 60℃,and ultrasonic power was 424 W,under these conditions,the yield of polysaccharide was 7.31%.In addition,this study was also conducted on the in vitro antioxidant capacity of polysaccharides from Tongqiao No.3.The results showed that the concentration of polysaccharide at 1.0 g/mL could eliminate DPPH radical and ABTS+radical capacity was 62.31%and 88.78%,and the FRAP was 76.76%.All of the results can provide a theoretical basis for the development and utilization of Tongqiao No.3.
