响应面法优化九头狮子草总黄酮提取工艺及其抗氧化活性研究 被引量:1

Study on optimization of extraction process of total flavonoids from Peristrophe japonica(Thunb.)Bremek.by response surface methodology and its antioxidant activity

摘要 以乙醇水溶液为溶剂,采用超声波辅助法提取九头狮子草总黄酮,在单因素试验基础上,通过响应面试验优化提取工艺,并对其DPPH·、ABTS^(+)·清除率及总抗氧化能力进行研究。结果表明:最佳提取工艺为乙醇体积分数30%、液料比50∶1(mL/g)、超声时间35 min,超声温度30℃,在此条件下总黄酮得率为1.60%,与理论预测值接近,相对标准偏差RSD为2.43%。当九头狮子草总黄酮质量浓度为100μg/mL时,DPPH·、ABTS^(+)·清除率分别为65.3%、74.1%,IC50为79.9、45.3μg/mL,而当其质量浓度为300μg/mL时,总抗氧化能力为0.476mmol/L,说明九头狮子草总黄酮具有一定的抗氧化性能。 Using ethanol aqueous solution as solvent,ultrasonic assisted extraction of total flavonoids from Peristrophe japonica(Thunb.)Bremek.was carried out.Based on the single factor experiment,the extraction process was optimized by response surface methodology.The DPPH.,ABTS^(+).clearance rate and total antioxidant capacity were studied.The results showed that the optimal extraction conditions were ethanol volume fraction 30%,liquid to material ratio 50:1(mL/g),ultrasound time 35 min and ultrasound temperature 30℃.Under these conditions,the yield of total flavonoids was 1.60%,which was close to the theoretical prediction value,and the relative standard deviation was 2.43%.When the mass concentration of total flavonoids from Peristrophe japonica(Thunb.)Bremek.was 100μg/mL,the DP­PH•and ABTS^(+)•scavenging rates were 65.3%and 74.1%,and IC50 were 79.9 and 45.3μg/mL,respectively.When the mass concentration of total flavonoids was 300μg/mL,the total antioxidant ca­pacity was 0.476 mmol/L.The results indicated that the total flavonoids of Peristrophe japonica(Thunb.)Bremek.had certain antioxidant properties.
