针刺对慢性疲劳模型大鼠海马TLR4/NF-κB信号通路及星形胶质细胞的影响 被引量:2

Effects of Acupuncture on TLR4/NF-κB Signaling Pathway and Astrocytes in Hippocampus of CFS Model Rats

摘要 目的:探究针刺对慢性疲劳模型大鼠海马TLR4/NF-κB信号通路及星形胶质细胞的影响。方法:将SD大鼠分为对照组、模型组与针刺组,模型组与针刺组进行慢性束缚造模,造模后针刺组进行针刺干预。对各组大鼠进行旷场实验观察大鼠行为学变化;RT-PCR检测Tlr4、Nfkb mRNA表达;Western blot检测TLR4、p-NF-κB/NF-κB、MyD88及GFAP蛋白表达;免疫组化技术检测GFAP表达情况;Sholl分析星形胶质细胞突起形态变化。结果:针刺可改善慢性疲劳综合征(CFS)大鼠的行为学变化;模型组Tlr4、Nfkb mRNA表达升高,针刺组降低;与对照组比较,模型组TLR4、p-NF-κB蛋白表达升高,针刺组与模型组比较TLR4、p-NF-κB蛋白表达降低,差异有统计学意义;与对照组比较,Western blot与免疫组化技术显示模型组GFAP蛋白表达下降,针刺组与模型组比较GFAP蛋白表达上调,差异有统计学意义;Sholl分析结果显示模型组星形胶质细胞突起受到抑制,针刺能够修复CFS引起的星形胶质细胞突起抑制。结论:针刺可以抑制CFS引起的神经炎症同时对星形胶质细胞起到调节作用,可能是其治疗CFS的机制之一。 Objective:To explore the effects of acupuncture on TLR4/NF-κB signaling pathway and astrocytes in hippocampus of chronic fatigue syndrome(CFS)model rats.Methods:SD rats were randomly divided into the control group,the model group and the acupuncture group.CFS model was established by chronic restraint.The acupuncture group was intervened with acupuncture.Behavior changes were observed in the groups by using open filed test.The mRNA expressions of Tlr4 and Nfkb were detected by RT-PCR.The protein expressions of TLR4,p-NF-κB/NF-κB,MyD88 and GFAP were detected by Western blot.The expression of GFAP was detected by immunohistochemistry and the morphological changes of astrocytes were analyzed by Sholl.Results:Acupuncture could improve the behavior of rats with CFS.The mRNA expressions of Tlr4 and Nfkb were increased in the model group,whereas they were decreased in the acupuncture group.Compared with those in the control group,the protein expressions of TLR4 and p-NF-κB were increased in the model group(P<0.01);compared with those in the model group,the protein expressions of TLR4 and p-NF-κB were decreased in the acupuncture group(P<0.01).The expression of GFAP was decreased in the model group compared with that in the control group(P<0.05),which was up-regulated in the acupuncture group(P<0.01).Sholl analysis showed that protuberances of astrocytes were inhibited in the model group,and acupuncture could repair the inhibition effect.Conclusion:Acupuncture can improve the CFS-induced neuroinflammation and astrocytes protuberances inhibition,which may be one of its mechanisms in treating CFS.
