电针不同穴位对束缚模型大鼠血清白细胞介素2及不同组织β内啡肽含量的影响 被引量:9

摘要 目的:观察电针对束缚模型大鼠血清白细胞介素2含量及血浆、下丘脑、垂体、淋巴结β内啡肽含量动态变化的影响,探讨免疫功能失调状态下白细胞介素2、β内啡肽含量变化的特点及二者的内在相关性。方法:实验于2004-01/04在北京中医药大学针灸学院针灸机理实验室完成。实验动物选取Wistar雄性大鼠40只。采用抽签分组方法,随机分为5组,正常对照组,束缚模型组,束缚针刺关元组,束缚针刺膻中组,束缚针刺非穴组,每组8只。制作束缚应激模型,分别选取“膻中”、“关元”、“关元旁开1.5寸非穴”进行电针刺激。膻中平刺0.2-0.3寸;关元及非穴均直刺0.5寸。针刺后接韩氏电针仪,断续波,频率2~100Hz,强度2~5mA,每次20min,隔日1次,共10次。采用放免法测定血清白细胞介素2含量及血浆、下丘脑、垂体、淋巴结β内啡肽含量并与正常组及模型组对照。并对血浆β内啡肽与血清白细胞介素2进行相关性分析,计量资料以x±s表示,各组组间差异采用方差分析,以P<0.05作为具有显著性差异的标准。结果:纳入40只大鼠均进入结果分析①各组大鼠血清白细胞介素2含量:束缚针刺膻中组明显高于正常对照组,束缚组,束缚针刺关元组犤(7.58±2.51),(5.42±1.12),(5.35±0.54),(5.33±0.92)ng/L,P<0.05犦。②各组大鼠血浆、下丘脑、垂体、淋巴结β内啡肽含量:束缚模型组,束缚针刺关元组,束缚针刺膻中组,束缚针刺非穴组血浆β内啡肽含量低于正常对照组,后三组高于束缚模型组高;后三组的组织β内啡肽(下丘脑、垂体、淋巴结)均比对照组高,比束缚组低(除外淋巴结β内啡肽)。③血清白细胞介素2与血浆β内啡肽的相关性:束缚针刺膻中组、束缚针刺关元组的大鼠血浆β内啡肽与血清白介素2呈正相关(r=0.67,0.58,P<0.05)。结论:针刺可提高束缚模型大鼠血清白细胞介素2的含量,降低垂体、下丘脑β内啡肽含量� AIM:To observe the influence of electro acupuncture on the dynamic changes of the content of interleukin-2 in serum and the contents of betaendorphin in plasma, hypothalami, pituitary gland and lymph node in adjuvant bondage stress model rats, and investigate the changing characteristics of interleukin-2 and beta-endorphin under status of immunological functional disorder and the inner association between them.METHODS:The experiment was carried out in the laboratory of acupuncture mechanism, School of Acupuncture, Beijing University of Traditional Chinese Medicine between March and June 2004. Forty male Wistar rats were randomly divided into 5 groups with 8 rats in each group: normal control group, bondage model group, bondage acupuncture at guanyuan,Shanzhong and non-point groups. Models of adjuvant bondage stress were established, and electroacupuncture was applied at Shanzhong and Guanyuan aeupoints, and the non-point which was 1.5 cun away from Guanyuan acupoint respectively; The Shanzhong acupoint was horizontally inserted to about 0.2 to 0.3 cun, and both Guanyuan acupoint and nonpoint were straightly inserted to about 0.5 cun. After acupuncture, Han electreacupuncture apparatus was connected, interrupted wave was given,the frequency was 2 to 100 Hz, the intensity was 2 to 5 mA, 20 minutes for each time, once every other day for a total of 10 times. Serum content of interleukin-2 and contents of beta-endorphin in plasma, hypothalami, pituitary gland and lymph node were detected with radioimmunoassay, and the results were compared with those in the normal control group and model group; The correlation between plasma beta-endorphin and serum interleukin-2 was analyzed, the data were expressed as mean±SD; Intergroup difference adopted analysis of variance, and P 〈 0.05 was taken as the standard for significant difference.RESULTS: All the 40 rats were involved in the analysis of results. ①Serum content of interleukin-2: It was obviously higher in the bondage acupuncture at shanzhong group than in
