摘要
目的探讨宁神安脏饮对肝郁脾虚型失眠大鼠行为学变化和下丘脑伽γ-氨基丁酸(GABA)及谷氨酸(GLU)表达的影响。方法采用夹尾刺激+饮食失联+PCPA多因素制备肝郁脾虚型失眠大鼠模型。将80只大鼠随机分为正常组、模型组、宁神安脏饮组和艾司唑仑组,正常组及模型组给予等体积生理盐水灌胃,宁神安脏饮组大鼠按每次2.28 g/kg灌胃给药,艾司唑仑组大鼠每次按剂量0.18 mg/kg灌胃给药,分别于实验第0、7、14、21天检测各组大鼠的体重,于第21天进行各组大鼠糖水偏好实验、旷场实验及Morris水迷宫实验,测试各组大鼠的行为学表现;应用透射电镜观察下丘脑组织结构的变化,免疫组织化学方法观察失眠21d后各组大鼠下丘脑γ-氨基丁酸(GABA)和谷氨酸(GLU)阳性细胞数。结果与正常组比较,模型组大鼠一般状态、体重、糖水消耗和糖水偏好率、旷场活动能力明显降低,水迷宫平均逃避潜伏期明显升高(P<0.05),透射电镜观察脑组织线粒体、内质网水肿;经给药干预后,与模型组比较,宁神安脏饮组和艾司唑仑组可改善大鼠的一般状态、体重、糖水消耗和糖水偏好率、旷场活动能力,降低水迷宫平均逃避潜伏期(P<0.05),透射电镜观察下丘脑组织结构明显改善。免疫组化结果表明,与正常组比较,模型组下丘脑GABA和GLU的阳性细胞数降低(P<0.05);与模型组比,宁神安脏饮组和艾司唑仑组GABA和GLU的阳性细胞数均升高(P<0.05)。结论宁神安脏饮可明显改善肝郁脾虚型大鼠的行为学,激发大鼠自发活动能力和探究能力,改善脑组织超微结构,其作用机制可能与调节失眠大鼠下丘脑GABA和GLU的含量有关。
Objective To investigate the effect of Ningshen Anzang Yin on behavioral changes and the expression of GABA and GLU in hypothalamus of insomnia rats with liver depression and spleen deficiency.Methods The rat model of insomnia with liver depression and spleen deficiency was established by clipping tail stimulation,diet disconnection plus PCPA.80 rats were randomly divided into normal group,model group,Ningshen Anzang Yin group and estazolam group.Normal group and model group was given equal volume of normal saline,Ningshen Anzang Yin group and estazolam group via lavage giving corresponding drugs,respectively in experimental 0,7,14,21 days of testing each rat body weight,sleep status,and on day 21,the behavioral performance of rats in each group was tested by sugar water preference test,open field test and Morris water maze test.The changes of hypothalamus tissue structure were observed by transmission electron microscope,and the contents ofγ-aminobutyric acid(GABA)and glutamate(GLU)in hypothalamus were observed by immunohistochemistry after 21 days of insomnia.Results Compared with the normal group,the general state,body weight,sugar water consumption,sugar water preference percentage and open field activity ability of rats in the model group were significantly decreased,the mean escape latency of water maze was significantly increased,and the hypothalamus neurons were severely damaged,with statistical significance(P<0.05).After drug intervention,compared with the model group,Ningshen Anzang Yin group and estazolam group increased the general state,body weight,sugar water consumption,sugar water preference percentage,opened field activity ability of rats,reduced the mean escape latency of water maze,and had a protective effect on hypothalamic neurons(P<0.05).The tissue structure of hypothalamus changed significantly under transmission electron microscope.Immunohistochemical results showed that comparing with normal group,the contents of GABA and GLU in model group were decreased(P<0.05).Compared with model grou
