摘要
目的:观察黄芩汤不同时间点的含药血清对TNF-α诱导的Caco-2细胞炎症模型的影响。方法:参照《伤寒论》组方及课题组的前期研究,煎煮、过滤、浓缩制成黄芩汤提取物,按照20 g/kg的剂量一次性给予大鼠黄芩汤,分别于给药后0、0.5、1、2、3、4、6、8、10、12、24 h眼眶取血,分离血清,MTT法观察不同浓度的黄芩汤含药血清对Caco-2细胞增殖的影响;并用TNF-α诱导细胞炎症损伤模型,10%的含药血清给药18 h后,采用Griess法观察黄芩汤含药血清对其NO的影响,采用Elisa法测定IL-1β、PGE_(2)和TNF-α的变化。结果:与正常组相比,5%、10%、20%的黄芩汤含药血清作用Caco-2后,均有促进其增殖的作用,尤以10%黄芩汤含药血清的效果最佳;10 ng/mL的TNF-α致炎效果最佳;不同时间点的10%黄芩汤含药血清可降低炎症细胞内NO的含量,降低细胞内IL-1β、TNF-α和PGE_(2)的浓度。结论:黄芩汤不同时间点的含药血清对TNF-α诱导的Caco-2细胞炎症具有明显的抑制作用,其作用机制可能与抑制NO、IL-1β、TNF-α等炎性因子有关。
Objective:To investigate the effect of Huangqin Decoction contained serum on Caco-2 cells induced by TNF-αat different time points.Methods:Referring to the formulation of Treatise on Febrile Diseases and the previous research,the extract of Huangqin Decoction was prepared after filtration and concentration,and then the extract was given to the rats(20 g/kg),the blood samples were collected at 0,0.5,1,2,3,4,6,8,10,12 and 24 h after administration,and the serum was separated.MTT method was used to detect the effect of different concentrations of Huangqin Decoction on Caco-2 cells proliferation.In addition,TNF-αwas used to induce cell inflammatory injury model.After incubated with 10%Huangqin Decoction contained serum for 18 hours,the content of nitric oxide(NO)was determined by Griess method and the concentrations of IL-1β,PGE_(2) and TNF-αwere measured by Elisa method.Results:Compared with the normal group,5%,10%,20%of Huangqin Decoction contained serum all had the effect of promoting proliferation,especially 10%of Huangqin Decoction contained serum had the best effect.TNF-αat 10 ng/mL had the best inflammatory effect.The concentrations of IL-1β,TNF-αand PGE_(2) in Caco-2 cells were decreased by 10%Huangqin Decoction contained serum at different time points.Conclusion:Huangqin Decoction contained serum at different time points has a significant inhibitory effect on TNF-αinduced inflammation of Caco-2 cells,and the mechanism may be related to the inhibition of NO,IL-1β,TNF-αand other inflammatory factors.
