新疆芜菁中均一多糖的分离纯化及结构分析 被引量:1

摘要 目的测定分离纯化后的芜菁多糖均一组分BP1和BP2含量及纯度,并对其进行结构分析。方法经DEAE-纤维素52和sephadex-G150柱层析分离纯化BP1和BP2,采用蒽酮-硫酸法在紫外-可见分光光度计进行纯度测定;采用比旋光度法、气相色谱法、傅里叶红外光谱法、核磁共振法等方法进行结构分析。结果BP1和BP2在分离得到的7种多糖组分中含量较高,纯度测定结果中在260 nm和280 nm处未见到蛋白质和核酸的吸收峰。BP1组分相对分子质量为110662,[α]为+18.83;BP2组分相对分子量为37844,[α]为+12.00。BP1、BP2均由阿拉伯糖(Ara)、甘露糖(Man)、葡萄糖(Glc)和半乳糖(Gal)组成,其摩尔比分别为为1∶2.90∶16.48∶1.52和1.24∶2.03∶15.24∶1。BP1主链由α-D-Man-(1→,→6)α-D-Glc-(1→跟→2,6和1→连接的β-D-Gal组成,α-D-Man-(1→,→6)α-D-Glc-(1→组成支链;BP2主链由→6)α-D-Ara-(1→,→6)α-D-Man-(1→和→6,1→)连接的β-D-Glc组成,→6和1→)连接的β-D-Glc组成支链。结论本研究初步成功推测出多糖均一组分BP1、BP2的结构特征并给后期芜菁多糖的结构鉴定提供了可靠的理论基础。 Objective To determine the content and purity of uniform content BP1 and BP2 polysaccharides from Brassica rapa L.,and to analyze the structures.Methods BP1 and BP2 wereseparated and purified by DEAE-cellulose 52 and Sephadex-G150 column chromatography,and thepurity were determined by Uv-Visible spectrophotometry and anthrone-sulfuric acid method.The molecular weights andthe specific rotation[α]were determined,and the structures were analyzed byGC,IR and NMR.Results The contents of BP1 and BP2 in the 7 components of polysaccharide were highestand did not show obvious absorption peaksin protein and nucleic acid at 260 nm and 280 nm after sephadex-g150isolated.Therelative molecular mass of component BP1 was 110662,the specific rotation was+18.83;The relative molecular massof component BP2 was 37844,and the specific rotation was+12.00.BP1 and BP2 were composed of Arabinose,Mannose,glucose and Galactose;the molar ratios were 1:2.90:16.48:1.52 and 1.24:2.03:15.24:1.Component BP1 backbone consistedα-D-Man-(1→,→6)α-D-Glcp-(1→and→2,6)β-D-Galp-(1→,α-D-)Man-(1→,→6)α-D-GlCp-(1→formsa branching chain;component BP2 backbone consists of→6)(α-D-Araf→,→6)α-D-Man-(1→and→6)β-D-Glcp-(1→,the branched chainconsists of→6)β-D-Glcp-(1→.)Conclusion In this study,the structural characteristics of Brassica rapa L.components BP1 and BP2 were predicted and reliable theoretical basis was provided for the later structural identification of Brassica rapa L.polysaccharides.
