摘要
采用水热法制备层状双氢氧化物(layered double hydroxide,LDH)纳米片,将改性聚乙二醇(H_(2)N-PEG-NH_(2))修饰在LDH表面,并进一步修饰靶向肽B3int,最后负载抗癌药物阿霉素(DOX)和光热剂吲哚菁绿(ICG),构建了一种新的DOX-ICG@LDH-PEG-B3int载药系统。通过扫描电子显微镜、紫外-可见分光光度计、热重分析仪等仪器对材料的形貌和理化性质进行表征,结果表明,DOX-ICG@LDH-PEG-B3int具有良好的形态分布,平均直径为77.68 nm。体外释药、体外细胞摄取、体外细胞毒性试验结果表明,DOX-ICG@LDH-PEG-B3int具有pH和近红外(NIR)双重刺激响应药物释放的性能,可以主动靶向肿瘤细胞,并可以显著提高DOX对细胞的杀伤作用以及ICG的光热效果。
Hydrothermal method was used to prepare layered double hydroxide(LDH)nanosheets.Modified polyethylene glycol(H_(2)N-PEG-NH_(2))was modified on the surface of LDH and further modified to the target peptide B3int.A new DOX-ICG@LDH-PEG-B3int drug delivery system was constructed by loading the anticancer drug doxorubicin(DOX)and photothermal agent indocyanine green(ICG).The morphology and physicochemical properties of the materials were characterized by scanning electron microscope,UV-vis spectrophotometer,thermogravimetric analyzer and other instruments.The results show that DOX-ICG@LDH-PEG-B3int has a good morphological distribution,with an average diameter of 77.68 nm.In vitro drug release,in vitro cell uptake and in vitro cytotoxicity tests indicate that DOX-ICG@LDH-PEG-B-3int has the pH and near-infrared(NIR)in response to drug release,which can actively target tumor cells,significantly improve the killing effect of DOX on cells and the photothermal effect of ICG.
