摘要
目的:观察芪仙通络方不同剂量对脑梗死大鼠血管新生及血管内皮生长因子(Vascular endothelial growth factor,VEGF)表达的影响。方法:大鼠随机分为假手术组、模型组、芪仙通络方15.66 g/kg、31.32g/kg、7.83 g/kg组和丁苯酞组,线栓法制备大脑中动脉阻塞大鼠模型,从造模后第3日起灌胃给药并同时腹腔内注射5-乙炔基-2'-脱氧尿苷(EDU)标记脑内增殖细胞,每日1次,连续12日。于造模后第3、7、14日对大鼠神经功能进行评分;免疫荧光染色观察大鼠缺血侧EDU/血小板内皮细胞黏附因子-1(platelet ndothe-lial cell adhensionmolecule-1,PECAM-1/CD31)双标阳性细胞数;免疫组化检测大鼠梗死区VEGF表达。Person相关分析法分析EDU/CD31双标阳性细胞数与神经功能评分和VEGF表达的相关性。结果:与假手术组比较,模型组大鼠各时间点神经功能评分均升高(P<0.05),大鼠缺血侧EDU/CD31双标阳性细胞数与VEGF表达升高,但二组差异无统计学意义(P>0.05);与模型组比较,芪仙通络方低剂量、中计量、高剂量组及丁苯酞组大鼠第7、14日神经功能评分明显降低(P<0.05),大鼠梗死区EDU/CD31双标阳性细胞数明显增多(P<0.05),VEGF表达明显升高(P<0.05),以高剂量组作用最为明显(P<0.05)。芪仙通络方高剂量组EDU/CD31双标阳性细胞数与神经功能评分呈显著负相关,与VEGF表达呈显著正相关。结论芪仙通络方可促进MCAO大鼠血管新生,其机制可能与上调缺血侧VEGF表达有关。
Objective To observe the effects of Qixian Tongluo Prescription on angiogenesis and VEGF expression in rats with cerebral infarction;To explore its mechanism.Methods The rats were randomly divided into shamoperation group,model group,Qixian Tongluo Prescription group formula 15.66 g/kg,31.32g/kg,7.83 g/kg groups and Butylphthalein group.The suture method was used to replicate the middle cerebral artery occlusion model in rats.The intragastric administration was started on the 3rd day after modeling,and 5-acetyl-2‘-deoxyuridine(EDU)was injected intraperitoneally to label proliferating cells in the brain once a day for 12 consecutive days.On the 3rd,7th,and 14th day after model establishment,neurological function was scored;immunofluorescence staining was used to observe ischemic side EDU/CD31 double-labeled positive cells;immunohistochemistry was used to detect the expression of Vascular endothelial growth factor(VEGF)in the ischemic side of rats;Pearson correlation analysis method was used to analyze the correlation between the number of EDU/CD31 double-labeled positive cells with neural function score and VEGF expression.Results Compared with the sham-operated group,the neurological function scores of rats in the model group were higher at all time points(P<0.05),and the number of EDU/CD31 positive cells and VEGF expression in the ischemic side of the rats were higher,but the difference between the two groups was not statistically significant(P>0.05);compared with the model group,the neurological function scores of rats in the low,medium and high dose groups of Qixian Tongluo Formula and the butylphthalide group were significantly lower on days 7 and 14(P<0.05).The number of EDU/CD31 positive cells in the infarct area increased significantly(P<0.05)and the expression of VEGF increased significantly(P<0.05),with the most obvious effect in the high-dose group(P<0.05).The number of EDU/CD31 double-labeled positive cells in the high-dose group of Qixian Tongluo Formula showed a significant negative correlation wit
