摘要
利用水蒸气蒸馏法提取新鲜珠兰叶中的挥发油,通过气相色谱-质谱联用技术(GC-MS)对其挥发油化学成分进行分析,采用1,1-二苯基-2-三硝基苯肼(DPPH)、2,2’,-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)、抑制酪氨酸酶法、抑菌圈法对挥发油的抗氧化和抑菌活性进行评价。结果表明:从珠兰叶挥发油中共检测出化合物55个,占挥发油总量的82.60%;其中以烯类化合物相对百分比最高(60.46%),其次是醇类化合物(14.52%),其主要成分为1,5,5-三甲基-6-亚甲基-环己烯(14.58%)、二环大根香叶烯(11.12%)、(4E,6Z)-2,6-二甲基-2,4,6-辛三烯(4.40%)、桉油烯醇(3.81%)、α-异黄樟烯(3.47%)、d-杜松烯(3.20%)等。珠兰叶挥发油对枯草芽孢杆菌、金黄色葡萄球菌、绿脓杆菌、大肠杆菌、变形杆菌均有显著的抑菌效果,对DPPH、ABTS自由基清除作用的抑制质量浓度(IC _(50))分别为14.112、3.766 g·L^(-1),对酪氨酸酶抑制作用的抑制质量浓度(IC _(50))为22.900 g·L^(-1),抗氧化活性及酪氨酸酶抑制作用与珠兰叶挥发油呈浓度效应。珠兰叶挥发油具有一定的抗氧化、酪氨酸酶抑制作用及抑菌特性。
The volatile oil was extracted by water-steam distillation and its chemical constituents were analyzed by GC-MS.The antioxidant activities of the volatile oil was determined by free radical scavening assays against 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2’-Azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS).Its antibacterial activities was also determined by inhibition zone.The inhibitory effect of the volatile oil on tyrosinase was studied.The fifty-five compounds were detected from the volatile oil of Chloranthus spicatus,accounting for 82.60%of the total.The content of alkene compounds was the most(60.46%),followed by alcohol compounds(14.52%).The major compounds of the volatile oil were 1,5,5-trimethyl-6-methylenecyclohexene(14.58%),bicyclogermacrene(11.12%),(4E,6Z)-2,6-dimethyl-2,4,6-octatriene(4.40%),spathulenol(3.81%),α-isosafrole(3.47%),d-cadinene(3.20%).The volatile oil also had the inhibition effects on Bacillus subtilis,Staphylococcus aureus,Pseudomonas aeruginosa,Escherichia coli and Bacillus proteus.When the scavenging rate against DPPH,ABTS free radicals and tyrosinase was 50%,the concentration of volatile oil were 14.112,3.766 and 22.900 g·L^(-1),respectively.The antioxidant activity and the inhibition of tyrosinase increased with the concentration of volatile oil.Therefore,the volatile oil from C.spicatus leaves showed a certain degree of antioxidant,antimicrobial activities and the inhibition of tyrosinase.
