HS-SPME-GC-MS analysis of volatile components in Scutellaria-Gardenia pair

摘要 目的分析比较黄芩-栀子药对及其单味药的挥发性成分。方法采用顶空固相微萃取(HSSPME)与气相色谱-质谱(GC-MS)联用的方法,测定黄芩-栀子药对及其单味药的挥发性成分,通过单因素试验,确定最佳萃取条件,采用面积归一法测定各成分含量。结果最终测定最佳条件为称量黄芩-栀子药对混合16目粗粉0.8 g,萃取温度100℃,平衡时间10 min,解析时间3 min。在药对及单味药中共检测出98种成分,在黄芩-栀子药对中鉴定出45种成分,占挥发性成分总量的97.59%;在黄芩中鉴定出50种成分,占挥发性成分总量的91.86%;在栀子中鉴定出40种成分,占挥发性成分总量的95.36%。结论黄芩和栀子配伍后,药对中挥发性成分的种类和含量较单味药相比在一定限度上发生了变化,两药配伍可能具有协同增效或减毒的作用,为后续研究包含黄芩-栀子药对的复方提供参考。 Objective To analyze and compare the volatile components of Scutellaria-Gardenia pair and its single compound.Methods Headspace solid phase microextraction(HS-SPME)and gas chromatography-mass spectrometry(GC-MS)were used to determine the volatile components of Scutellaria-Gardenia pair and its single compounds.The optimal extraction conditions were determined by single factor test,and the content of each component was determined by area normalization method.Results The optimal conditions for the final determination were as follows:weighing of Scutellaria-Gardenia pair mixed coarse powder 0.8 g,extraction temperature 100℃,equilibrium time 10 min,and analytical time 3 min.A total of 98 ingredients were detected in drug pair and single drug.A total of 45 components were identified in Scutellaria-Gardenia pair,accounting for 97.59%of the total volatile components.A total of 50 components were identified in Scutellaria,accounting for 91.86%of the total volatile components.A total of 40 components were identified from Gardenia,accounting for 95.36%of the total volatile components.Conclusion After the compatibility of Scutellaria and Gardenia,the types and contents of volatile components in the drug pairs changed to a certain extent compared with that of single drug.The compatibility of the two drugs may have synergistic effect or toxicity reduction,which provides reference for the subsequent study of the compound containing scutellaria and gardenia.
