石菖蒲对脑梗死大鼠认知功能障碍及神经细胞凋亡的影响及机制探讨 被引量:18

Effect and Mechanism of Acorus Gramineus Soland on Cognitive Dysfunction and Neuron Apoptosis in Rats with Cerebral Infarction

摘要 目的分析石菖蒲对脑梗死大鼠认知功能障碍及神经细胞凋亡的影响并探讨其作用机制。方法将60只大鼠随机分为假手术组、阳性对照组、模型组、石菖蒲低剂量组、石菖蒲高剂量组。采用线栓法阻断大脑中动脉建立脑梗死模型,其中假手术组不栓塞大脑中动脉。建模成功后,石菖蒲低剂量组(10只)、石菖蒲高剂量组(11只)分别以石菖蒲0.6 g/kg、1.8 g/kg灌胃,阳性对照组(11只)以尼莫地平1 mg/kg灌胃,假手术组(11只)和模型组(10只)灌胃生理盐水,每天1次,连续干预30 d。干预完成后观察大鼠的行为学状态,水迷宫实验评价大鼠认知功能,Tunel染色观察梗死周边组织神经细胞凋亡情况,实时荧光定量聚合酶链式反应(PCR)检测梗死周边组织糖原合成酶激酶-3β(GSK-3β)、β-连环蛋白(β-catenin)、促凋亡基因(Bax)、半胱天冬酶3(Casp3)mRNA相对表达量,蛋白免疫印迹法(Western Blot)检测GSK-3β、磷酸化GSK-3β(p-GSK-3β)、β-catenin、p-β-catenin、Bax、Casp3蛋白相对表达量。结果模型组大鼠精神状态较差,活动量减少,有明显的行走时向对侧倾斜、旋转追尾、竖毛、瘫痪等神经功能缺损表现;石菖蒲高剂量组有轻度对侧肌张力升高、竖毛、身体向对侧倾斜等表现,神经功能损伤较石菖蒲低剂量组轻。与假手术组比较,模型组逃避潜伏期延长、神经细胞凋亡数更多,穿越平台次数与目标象限停留时间百分比均减少(P<0.05);与模型组比较,阳性对照组及石菖蒲干预组逃避潜伏期更短、神经细胞凋亡数更少,穿越平台次数与目标象限停留时间百分比增加(P<0.05);与阳性对照组和石菖蒲低剂量组比较,石菖蒲高剂量组逃避潜伏期更短,神经细胞凋亡数更少,穿越平台次数与目标象限停留时间百分比增加(P<0.05)。与假手术组比较,模型组p-GSK-3β/GSK-3β、p-β-catenin/β-catenin比值及Bax、Casp3 mRNA和蛋白相对表达量更高(P<0.05);与 Objective To analyze the effect of acorus gramineus soland on cognitive dysfunction and neuron apoptosis in rats with cerebral infarction and explore its mechanism.Methods Sixty rats randomly divided into sham operation group,positive control group,model group,acorus gramineus soland low-dose group and acorus gramineus soland high-dose group.The middle cerebral artery was blocked by thread embolization method to establish the cerebral infarction model,in which the middle cerebral artery was not embolized in the sham operation group.After the model established successfully,the acorus gramineus soland low-dose group(10 rats)and acorus gramineus soland high-dose group(11 rats)were gavaged with 0.6 g/kg and 1.8 g/kg body weight of acorus gramineus soland,respectively.The positive control group(11 rats)were gavaged with nimodipine 1 mg/kg body weight.The sham operation group(11 rats)and the model group(10 rats)were gavaged with normal saline,once a day,for 30 days of continuous intervention.After the intervention,the behaviors of rats were observed.The cognitive function of rats was evaluated by water maze.Tunel staining was used to observe the apoptosis of nerve cells around the infarct tion.Real-time fluorescence quantitative PCR was used to detect the expression of glycogen synthase kinase-3β(GSK-3β),β-Catenin,B apoptosis cell matrixgene(Bax),and caspase-3(Casp3)mRNA.Western blot was used to detect the relative expression of GSK-3β,phosphorylated(p)-GSK-3β,β-Catenin,p-β-Catenin,Bax,and Casp3.Results The rats in the model group with poor mental state,activity decreased,and there were obvious neurological deficits such as tilting to the opposite side,rotating rear-end,with vertical hair,and paralysis when walking.The acorus gramineus soland high-dose group shared mild contralateral muscle tension,vertical hair,the body tilted to the opposite side,and the neurological impairment was lighter than that in the acorus gramineus soland low-dose group.Compared with the sham operation group,the escape latency was prol
