摘要
以体长(1.932±0.204)cm、体质量(1.386±0.055)g的1月龄棘胸蛙蝌蚪为研究对象,以尾部皮肤ATP酶、SOD酶、CAT酶和肝脏SOD酶、CAT酶、GSH酶活力为测定指标,依次设置5.5、6.5、7.0、7.5、8.5和9.5等6个pH实验梯度,在水源水温(24±0.2)℃、DO(7.30±0.01)mg·^(L-1)、pH(7.30±0.01)条件下,开展了棘胸蛙蝌蚪对pH急性胁迫阶段(实验时长96 h,测定时点依次为24、48、72、96 h)及恢复阶段(恢复至pH 7.0,实验时长12 d,测定时点分别为6 d和12 d)的抗氧化应答特征研究。结果表明:(1)pH急性胁迫阶段,各测定时点尾部皮肤ATP酶和肝脏GSH酶活力均随pH的升高呈先降后升趋势,皮肤和肝脏表露SOD和CAT酶活力均随pH的升高呈先升后降趋势,其中pH 7.5为上述酶活力值发生转折的分水岭;(2)pH恢复阶段,各实验组的恢复6 d和12 d的酶活力均无显著差异(P>0.05);以各实验组pH急性胁迫96 h时点的酶活力为参照,除pH 7.0和7.5两实验组的尾部皮肤ATP、SOD、CAT酶活力和肝脏SOD、CAT、GSH-Px酶活力均与其相对应的参照组无显著差异(P>0.05)外,其余实验组均显著大于与其相对应的参照组(P<0.05),但尾部皮肤ATP酶和肝脏GSH-Px酶活力均仍持续显著大于pH 7.5实验组(P<0.05),尾部皮肤和肝脏的SOD、CAT酶活力均仍持续显著小于pH 7.5实验组(P<0.05)。
Using 1-month-old Quasipaa spinosa tadpoles of(1.932±0.204)cm body length and(1.386±0.055)g body weight as the experimental animal,tail skin′s ATP,SOD,CAT activities and liver′s SOD,CAT,GSH activities of Q.spinosa tadpole as measurement indicators,setting up 6 pH experimental gradients of 5.5,6.5,7.0,7.5,8.5 and 9.5 in sequence,and under the conditions of source water temperature(24±0.2)℃,DO(7.30±0.01)mg·L^(-1),and pH(7.30±0.01),studied the characteristics of anti-oxidant response on the stage of acute pH stress(experimental duration was 96 h,measurement time points were 24,48,72,96 h)and recovery(recovery to pH 7.0,experiment duration was 12 d,measurement time points were 6 d and 12 d).The results showed that:(1)During the acute challenge stage,the ATPase activity of tail skin and GSH enzyme activity of liver decreased first and then increased with the increment of the pH,the SOD and CAT enzyme activities of the skin and liver increased first and then decreased with the increment of the pH,among which the pH 7.5 was the turning point of the above enzyme activities value.(2)During the recovery stage,there was no significant difference in the enzyme activity of each experimental group at 6 d and 12 d(P>0.05);Compared with the enzyme activity at 96 h in the pH challenge stage of each experimental group,except that there was no significant difference(P>0.05)with showed that the tail skin ATP,SOD,CAT enzyme activities and the liver SOD,CAT,GSH-Px enzyme activities in the pH 7.0 and 7.5 experimental groups,the enzyme activities of other experimental groups were significantly greater than that of the corresponding group,all other experimental groups(P<0.05).However,the activities of tail skin ATPase and liver GSH-Px enzyme activities were always significantly greater than the pH 7.5 experimental groups(P<0.05),and the SOD and CAT enzyme activities of the tail skin and liver were always significantly lower than the pH 7.5 experimental group(P<0.05).
