3种中草药对悬游双眉虫抗氧化酶活性的影响

摘要 为探究3种中草药(槟榔、土槿皮、仙鹤草)对悬游双眉虫的抗氧化酶活性影响,本研究以悬游双眉虫为实验对象,实验生态条件下采用不同浓度中草药提取液进行杀灭试验,研究了悬游双眉虫的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)以及谷胱甘肽过氧化物酶(GPx)活性的剂量-效应关系和时间-效应关系,利用整合生物标志物响应(IBR)指数评价3种中草药对悬游双眉虫的毒性效应。结果显示:悬游双眉虫在3种中草药的作用下的3种抗氧化酶活性的剂量-效应关系呈现先上升后下降的趋势(倒U型曲线);槟榔、土槿皮作用下的IBR值总体上呈现先升后降的趋势,仙鹤草作用下IBR值则先下降后上升。根据IBR值大小分析得到三种中草药对悬游双眉虫抗氧化酶活性影响大小依次为:仙鹤草>土槿皮>槟榔,CAT和GPx在药物暴露时间和浓度变化下均呈现了极显著差异(P<0.01),因此,IBR指数与CAT、GPx结合可作为有效的生物标志物。 In order to explore the effects of Chinese herbal medicines on the antioxidant enzyme activities of harmful ciliates in aquaculture,the effects of toxicity by Areca catechu,Cortex Pseudolaricis,Agrimonia pilosa on superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GPx)of Diophry sappendiculata were studied.Integrated biomarker responses index(IBR)method was used to quantitatively assess toxic effects of three Chinese herbal medicines on D.appendiculata.The results illustrated that the dose-response relationship of the antioxidant enzyme activities under the action of three Chinese herbal medicines firstly increased and then decreased.The time-response relationship under the treatment of A.catechu also showed a trend of increasing at first and then decreasing;in addition,the CAT activities under the treatment of C.Pseudolaricis decreased with time,while the CAT and GPx activities under the treatment of A.pilosa decreased with time.Furthermore,under the action of A.catechu and C.Pseudolaricis,the value of IBR first increased and then decreased,while under the action of A.pilosa,the value of IBR first decreased and then increased.Additionally,according to the analysis of IBR value,the effects of three Chinese herbal medicine to D.appendiculata in the descending order were as follows:A.pilosa,C.Pseudolaricis,A.catechu.Moreover,Glutathione peroxidase(GPx)and catalase(CAT)showed extremely significant differences(P<0.01).Therefore,IBR index combined with CAT and GPx can be used as an effective biomarker.
