安肠汤对正虚邪恋型溃疡性结肠炎大鼠miRNA-146a/IRAK-1/NF-κB信号通路的影响 被引量:10

摘要 目的观察安肠汤对对正虚邪恋型溃疡性结肠炎大鼠miRNA-146a/IRAK-1/NF-κB信号通路的影响,探讨安肠汤干预溃疡性结肠炎的可能作用机制。方法随机选取SPF级SD大鼠15只作为空白组,另取45只大鼠采用2,4,6-三硝基苯磺酸/乙醇灌肠法建立溃疡性结肠炎模型,将造模成功大鼠随机分为模型组、丽珠肠乐组、安肠汤组各15只。空白组、模型组灌胃生理盐水,丽珠肠乐组灌胃丽珠肠乐混悬液,安肠汤组灌胃安肠汤。灌胃14 d后,HE染色观察大鼠结肠病理组织学改变,通过定量聚合酶链反应(qPCR)测定大鼠血浆中miRNA-146a表达水平,Western blot法检测结肠组织中IRAK-1、NF-κB蛋白表达水平,ELISA法检测血清中白细胞介素-10(IL-10)、白细胞介素-17(IL-17)、肿瘤坏死因子-α(TNF-α)水平。结果模型组大鼠肠黏膜表面出现缺损,大量炎细胞聚集、浸润,腺体破坏甚至丢失;丽珠肠乐组和安肠汤组大鼠肠黏膜缺损、腺体破坏较模型组轻,炎细胞较模型组少,安肠汤组病变更轻。模型组大鼠血浆中miRNA-146a相对表达量,结肠上皮组织中IRAK-1、NF-κB蛋白表达量和血清中IL-17、TNF-α水平均明显高于空白组(P均<0.05);丽珠肠乐组和安肠汤组上述各指标均明显低于模型组(P均<0.05);安肠汤组血浆中miRNA-146a相对表达量和结肠上皮组织中IRAK-1、NF-κB蛋白表达量均明显低于丽珠肠乐组(P均<0.05),血清中IL-17、TNF-α水平均明显高于丽珠肠乐组(P均<0.05);模型组、丽珠肠乐组和安肠汤组大鼠血清中IL-10水平均明显高于空白组(P均<0.05),丽珠肠乐组和安肠汤组均明显高于模型组(P均<0.05),安肠汤组明显高于丽珠肠乐组(P<0.05)。结论安肠汤可通过调控miRNA-146a/IRAK-1/NF-κB信号通路抑制炎症因子的表达来减轻肠黏膜炎症反应,从而起到治疗正虚邪恋型溃疡性结肠炎的作用。 Objective It is to observe the effect of Anchang Decoction on miRNA-146a/IRAK-1/NF-κB signaling pathway in rats with ulcerative colitis of body resistance weakened while pathogenic factors protracted,to explore the possible mechanism of Anchang Decoction in intervening ulcerative colitis.Methods Fifteen SPF SD rats were randomly selected as the blank group,and another 45 rats were used to establish the models of ulcerative colitis with 2,4,6-trinitrobenzenesulfonic acid/ethanol by enema.The successfully modeled rats were randomly divided into model group,Livzon bifidobacterium preparation group,and Anchang Decoction group,with 15 rats in each group.The blank group and the model group were given normal saline by gavage,the Livzon bifidobacterium preparation group was given Livzon bifidobacterium preparation suspension by gavage,and the Anchang Decoction group was given Anchang Decoction by gavage.After intragastric administration for 14 days,the histopathological changes of rat colon was observed by HE staining.The expression level of miRNA-146a in rat plasma was measured by quantitative polymerase chain reaction(qPCR),and the protein expression levels of IRAK-1 and NF-κB in colon tissue were detected by Western blot,the serum levels of interleukin-10(IL-10),interleukin-17(IL-17),tumor necrosis factor-α(TNF-α)were detected by ELISA method.Results The intestinal mucosal surface of rats in the model group had defects,with a large number of inflammatory cells aggregated and infiltrated and the glands destroyed or even lost;the intestinal mucosal defects and gland damage of the rats in the Livzon bifidobacterium preparation group and the Anchang Decoction group were lighter and the inflammatory cells were less than that in the model group,the pathological changes in the Anchang Decoction group were more lighter.The relative expression of miRNA-146a in plasma,the expression of IRAK-1 and NF-κB protein in colonic epithelial tissue,and the levels of IL-17 and TNF-αin serum in the model group were significantly high
